摘  要:目的:通过大鼠行为学评价慢性束缚应激致焦虑大鼠模型,观察逍遥散对焦虑模型大鼠的调节作用。方法:36只SD健康雄性大鼠,随机分为4组:正常组、模型组、逍遥散组和氟西汀组,每组9只。除正常组外,均给予14d慢性束缚应激建立模型。正常组和模型组大鼠给予等量去离子水灌胃,而逍遥散组和氟西汀组大鼠分别给予逍遥散(0.61g干粉/100g大鼠/d)以及氟西汀(0.2mg/100g大鼠/d)灌胃,每日1次,共14d。观察大鼠体质量变化及行为学指标,行为学包括糖水消耗实验、旷场实验、高架十字迷宫以及新环境进食抑制实验。结果:各组大鼠实验前进行体质量及行为学观察,经统计学处理无统计学差异,第15天,再次进行体质量及行为学观察,①大鼠体质量,模型组体质量明显低于其他3组(P<0.05),逍遥散组、氟西汀组与正常组比较差异无统计学意义;②糖水消耗实验,各组大鼠比较糖水消耗量无统计学差异;③旷场实验,各组大鼠运动总距离、中央区进入次数、中央区停留时间差异均无统计学意义;④高架十字迷宫实验,模型组大鼠运动总距离、双侧开放臂进入次数、双侧开放臂停留时间、中央区进入次数明显低于正常组(P<0.05,P<0.01),而双侧封闭臂进入次数、双侧封闭臂停留时间、中央区运动距离无统计学差异;逍遥散组、氟西汀组与正常组比较无统计学差异;⑤新环境进食抑制实验,模型组与正常组比较,进食时间明显较长(P<0.05),而逍遥散组、氟西汀组大鼠进食时间与正常组比较无统计学差异。结论:慢性束缚应激14d可致大鼠焦虑,逍遥散具有调节大鼠焦虑样行为作用。Objective: To evaluate anxiety rat model caused by chronic immobilization stress through behavioristics, and observe regulatory effects of Xiaoyaosan on anxiety rats. Methods: 36 male SD rats were randomly divided into 4 groups: normal group, model group, Xiaoyaosan group and fluoxetine group. Except normal group, other 3 groups were made anxiety model by 14 days chronic immobilization stress. Rats in normal and model groups were intragastric administered equivalent deionized water, while Xiaoyaosan group and fluoxetine group were gave corresponding drugs respectively, once a day, total 14 days. In 15th day, rat weights and behaviors were observed. Behavior experiments included glucose consumption, open-field, elevated plus maze and novelty suppressed feeding tests. Results: All group rats were conducted body weight and behavior tests before experiment, and there were no differences among them. In 15th day, (1)body weights of model group were lower than the other groups (P〈0.05); (2)there were no differences among all four groups in glucose consumption test; (3)open-field test showed no differences in all groups in terms of total movement distance, central zone entrance times and retention times; (4)elevated plus maze test showed total movement distance, open-arm entrance times, retention time and central zone movement in model group were lower than the other groups (P〈0.05, P〈0.01), while there were no statistical differences among other groups; (5)novelty suppression feeding test indicated feeding latent periods in model group were longer than others (P〈0.05). Conclusion: 14 days chronic immobilization stress can cause rat anxiety, and Xiaoyaosan has regulatory effects on the anxiety-like behaviors.
