摘　　要：目的:探讨中药注射液"喘可治"改善猴免疫缺陷病毒(SIV)感染的淋巴组织纤维化的作用机制及治疗价值。方法:将12只SIV感染的中国恒河猴随机分为2组,分别给予喘可治和生理盐水,持续给药2个月,定期采集血样和淋巴结,通过免疫组化、流式等技术检测相关指标变化。结果:喘可治可保护和重建感染SIV猴的淋巴结滤泡和T细胞区,使胶原沉积减少,网状组织结构增加并趋于有序,且此变化与TGF-β通路相关。结论:"喘可治"可重建淋巴组织网状结构,据此有望研发出靶向于免疫重建的新治疗方案。Objective： To investigate the mechanism and therapeutic value of TCM injection Chuankezhi （CKZ） on lymphatic tissue fibrosis in chronic simian immunodeficiency virus （SIV） infection. Methods： 12 Chinese rhesus macaques in SIV infection were assigned randomly into a treatment group （n=6） and a control group （n=6）, and received daily intramuscular injection of CKZ, and normal saline respectively for 2 consecutive months. Blood sample and lymph node biopsies were collected at the time-points as intialed. Parameters involving the fibrosis were determined by pathological and immunohistochemical stain, and flow cytometry. Results： CKZ treatment preserved and restored the network architecture of lymph node in both follicle and T cell zone, coincided with increased T cell proliferation as verified by Ki67 staining. We further observed that CKZ treatment reduced the collagen deposition, increased the desmin expression, and restored the desrnin positive reticular network, accompanied with TGF-β expression decreased. Conclusion： CKZ treatment reduces collagen deposition and improves the lymphatic network architecture through a TGF-β associated pathway. It can restore the lymphoid network architecture in SIV infected monkeys and support a novel therapeutic strategy for targeting CD4 reconstitution.
