硫酸铜抑制人卵巢癌腹水和新生血管生成的实验研究

摘  要:目的探讨水通道蛋白3(AQP3)抑制剂硫酸铜抑制人卵巢癌腹水和新生血管生成的机制。方法 21只裸鼠注射荷人卵巢癌乳头状腺癌细胞(SKOV3)建模后,分别腹腔注射硫酸铜5μg/g(A组)、Avastin 5μg/g(B组)和生理盐水1ml(C组)。每周一、二、四、五腹腔注射1次,共4周。检测腹水量、腹膜渗透性、微血管密度(MVD);应用Western blot法检测AQP3和血管内皮生长因子(VEGF)在腹水和卵巢癌组织中的表达。结果 A、B组腹水量少于C组(P<0.05);A组腹水及卵巢癌组织中的AQP3低于B、C组(P<0.05);A、B组腹水及卵巢癌组织中的VEGF低于C组[(206.18±16.47)ng/L、(192.88±14.31)ng/L vs.(408.37±18.41)ng/L](P<0.05)。AQP3及VEGF的表达与腹水量、腹膜渗透性及MVD呈正相关(P<0.01)。AQP3的表达与VEGF的表达呈正相关(P<0.01)。结论硫酸铜可能通过下调肿瘤组织中VEGF的表达抑制人卵巢癌裸鼠腹水的形成及新生血管的生长。Objective To investigate the mechanism of inhibitory effect of copper sulfate[an inhibitor of aquaporin 3(AQP3)] on human ovarian cancer-induced ascites and angiogenesis. Methods After the nude mice models with bearing human ovarian cancer papillary adenocarcinoma cells (SKOV3) were established, 21 model mice were injected copper sulfate 5μg/g(group A, 7 mice), Avastin 5 μg/g(group B, 7 mice) and normal saline 1 ml(group C, 7 mice). The injection was performed intraperitoneally on Monday, Tuesday, Thursday and Friday, once a day for 4 weeks. The amount of ascites, peritoneal permeability and microvessel density (MVD) were determined. The expressions of AQP3 and vascular endothelial growth factor(VEGF) in ascites and ovarian cancer were detected by Western blot. Results The amount of ascites was less in groups of A and B than that in group C(P〈0. 05). AQP3 expression in ascites and ovarian cancer was lower in group A than that in groups of B and C(P〈0. 05). The expressions of AQP3 and VEGF were positively correlated with the amount of ascites, peritoneal permeability and MVD(P〈0. 01). The expressions of AQP3 was positively correlated with that of VEGF(P〈0. 01). Conclusion Copper sulfate may inhibit human ovarian cancer-induced ascites and angiogenesis in nude mice, which may be related to the downregulation of VEGF expression in tumor tissues.
