摘  要:目的基于Kelch样ECH相关蛋白1(Keap1)-NF-E2相关因子-2/抗氧化反应元件(Nrf2/ARE)探讨干燥综合征(SS)模型大鼠心功能变化的机制。方法 30只Wistar雄性大鼠随机分为正常对照组、模型对照组,每组15只。模型组大鼠两后足的足跖部皮下注射完全弗氏佐剂联合同种鼠颌下腺抗原造模,诱导SS模型。致炎30 d后观察大鼠的体质量变化、饮水量变化、颌下腺指数、脾指数、腺体的组织学变化;有创血流动力学监测SS大鼠心功能的变化;ELISA检测血清中活性氧(ROS)、丙二醛(MDA)、超氧化物歧化酶(SOD)、总抗氧化能力(TAC)、IL-18、IL-35;免疫组织化学方法检测ROS、活性氮自由基(RNS)、谷胱甘肽(GSH)、硫氧还蛋白(TrX)蛋白表达情况;实时荧光定量PCR(qRT-PCR)检测Keap1、Nrf2、巨噬细胞活化因子(Maf)、ARE mRNA的表达;Western blot法测定大鼠心肌组织γ-谷氨酰半胱氨酸合成酶(γ-GCS)、血红素氧合酶1(HO-1)蛋白表达水平。结果与正常组比较,模型组大鼠饮水量变化显著升高(P<0.01),颌下腺指数、脾指数及体质量显著下降(P<0.05);模型组大鼠心率(HR)、心脏指数(HI)、左室收缩期压(LVSP)、左室舒张期压(LVEDP)显著升高(P<0.05),左室内压上升下降最大速率(±dp/dtmax)显著下降(P<0.05);模型组IL-18、MDA、RNS、TAC、ROS含量均升高,而TrX、GSH、IL-35、SOD含量下降;同时,心组织Keap1、Maf、Nfr2的mRNA,HO-1、γ-GCS蛋白表达水平亦上升(P<0.01)。结论 SS大鼠免疫平衡失调可能与Keap1、Maf、Nfr2的mRNA,HO-1、γ-GCS表达上调有关。Objective To investigate the mechanism underlying the changes in the cardiac function in rat models of Sj6gren' s syndrome (SS) based on Kelch-like ECH-associated protein 1 (Keapt)-Nrf2/ARE signaling pathways. Methods Thirty male Wistar rats were randomly divided into two groups, normal control (NC) group and SS model group, with 15 in each. The rats in the model group were injected with the complete Freund's adjuvant plus homologous antigen of submandibular gland (0.2 mL mixture) into bilateral posterior paw metatarsus. The body mass, water intake, submandibular gland index, spleen index, and histological changes of the glands were observed 30 days after inflammation was induced. Cardiac function was assessed using invasive hemodynamic monitoring. Meanwhile, reactive oxygen species (ROS), malondialdehyde (MDA), superoxide dismutase (SOD) and total antioxidant capacity (TAC), IL-18, IL-35 were detected using ELISA; ROS and reactive nitrogen species (RNS), glutathione (GSH), thioredoxin (TrX) protein were determined using immunohistochemical staining. The expressions of Keap 1, nuclear factor erythroid 2-related factor (Nrf2), macrophage activating factor (Maf) antioxidant responsive element (ARE) mRNAs were detected using real-time fluorescent quantitative PCR (qRT-PCR) ; the levels of heme oxygenase-1 (HO-1), γ-glutamic acid and a half long glycine synthetase (γ-GCS) proteins in cardiac tissue were examined using Western blotting. Results Compared with the NC group, the model group presented reduced body mass, submandibular gland index and spleen index (P 〈 0.05), increased water intake (P 〈 0.01 ), heart rate ( HR), heart index (HI), left ventricular systolic pressure (LVSP) and left ventricular diastolic pressure (LVEDP) ( P〈0.0.5), and decreased left ventricular ± dp/dtmax (P 〈0.05). Compared with the NC group, the model group had increased IL-18, MDA, RNS, TAC, ROS levels, and Keap1, Maf, Nfr2 m
