摘  要:研究开发适用于牛革、绵羊革、山羊革、马革的荧光PCR鉴定方法,优化了皮革DNA提取方法。该方法对皮革制品的DNA检出限为1ng DNA,具有很好的特异性。样品检测试验表明:方法稳定性强,优于已有检测方法,适用于动物性皮革制品的鉴别,对于甄别市面上动物性纤维制品的制假造假现象,规范动物性纤维制品产品质量,提供了技术支撑。DNA extraction and real - time PCR methods were developed for the identification of cow leather, sheep leather, goat leather and horse leather. Specificity experiment shows that all the four kinds of leather can be distinguished from other leath- er products using the species - specific probes, and sensitivity experiment demonstrates that the detection limit of the established method is 1 ng DNA. Based on the sample analysis result of fifteen leather products, the established methods are more reliable and sensitive than the traditional methods, and suitable for the routine detection of animal leather products, which will contribute to the discrimination of leather counterfeit phenomenon.
