摘 要:以丁二酸酐对聚乙烯醇进行改性制备改性聚乙烯醇,使其在EDC/NHS交联反应体系下与胶原共价交联。然后,再采用反复冻融物理交联的方式予以强化,制备化学交联-互穿网络共存的胶原基复合水凝胶。为了更好地掌握过程中各因素对交联反应的影响,采用响应曲面设计方法研究了改性聚乙烯醇中的羧基含量、改性聚乙烯醇与胶原质量比、交联剂用量以及冻融循环次数对水凝胶力学性能的影响。结果表明,响应面模型可以很好地拟合试验条件下所得到的各响应值。与各单因素相比,各平方项对水凝胶力学性能的影响更为明显。然而,仍需进一步对其进行结构、化学性能、生物学性能及其体表创伤修复效果等方面的评价。A novel covalent cross - linked polyvinyl alcohol - collagen hydrogel was prepared and the physical properties were studied using a response surface methodology. The carboxyl groups were introduced into PVA through the reaction with succinic anhydride. The EDC/NHS was used as cross - linker. The hydrogels were prepared by repeated freezing and thawing. For a better understanding of the influence of the parameters and their interaction, four factors such as the content of carboxyl groups, the ratio of modified PVA to collagen, the dosage of cross - linker and the cycles of freezing and thawing were investigated by a response surface methodology. The physical properties of the hydrogels ( tensile strength, elongation at break) were analyzed. The experiments with real data indicated that the response surface models could significantly fit the response values. Moreover, comparing with the single factor, the interactions between factors had more obvious effects on the physical properties of PVA - Collagen bydrogels. After the analysis of factors' influences and investigation of the data and trends, the structure of the hydrogels might bethe driving force of the different physical behaviors.
