摘  要:目的:通过研究瘦素基因敲除肥胖小鼠,探讨肥胖对肺组织炎症和纤维化的影响。方法:ob/ob基因敲除C57小鼠为肥胖组(8只),野生型C57小鼠为正常组(8只),喂养至5个月。分别采用苏木精伊红(hematoxylin-eosin,HE)染色观察肺组织炎症细胞的浸润,masson染色及人α平滑肌肌动蛋白(alpha smooth muscle actin,α-SMA)免疫组化观察支气管周围胶原纤维沉积及Western blot检测肺组织MMP-9的表达。结果:(1)肥胖组小鼠支气管周围有明显的炎症细胞浸润,较正常组明显增加(P=0.015),而肺泡周围则增加并不明显(P=0.266)。(2)肥胖组小鼠肺组织masson染色及α-SMA免疫组化支气管周围胶原纤维沉积较正常组均明显升高(α-SMA:P=0.003;masson:P=0.002)。(3)肥胖组小鼠肺组织中基质金属蛋白酶9(Matrix metalloproteinases 9,MMP-9)蛋白表达与正常组相比明显升高(P=0.006)。结论:通过对ob/ob基因敲除肥胖小鼠研究,推测肥胖可增加肺组织炎症及肺组织纤维化程度。Objective : To explore the effect of obesity on lung inflammation and fibrosis observed in leptin gene (ob/ob) knockout obesity mice. Methods : C57 mice of ob/ob gene knockout as obese group, eight wild-type C57 mice as normal group, feeding to 5 months. With hematoxylin-eosin staining to observe the infiltration of inflammatory cells in lung tissue, the masson staining and the immunohistochemieal of alpha smooth muscle actin were used to detect the deposition of collagen around bronchial, and Western blot was used to detect the expression of matrix metalloproteinase 9 ( MMP-9 ) in lung tissue respectively. Results : ( 1 ) The infiltration of inflammatory cells in lung tissue were obviously increased in obesity mice compared to normal nlice( P = 0. 015 ) , but there was no sig- nificant around alveolar( P = 0. 266 ). (2) Masson staining and immunohistochemical of a-SMA were shown that the collagen deposi- tion around bronchial in obesity mice significantly higher than normal mice ( c^-SMA : P = 0. 003 ; masson : P = 0. 002 ). ( 3 ) The expres- sion of MMP-9 in lung tissue was significantly increased in obesity mouse than normal mouse (P = 0. 006). Conclusion: We speculate obesity may increase inflammation and the degree of lung tissue fibrosis through the experiment to ob/ob gene knockout obesity mice.
