摘  要:目的利用基因芯片研究薯蓣皂苷元对人胃低分化粘液腺癌MGC-803细胞基因表达的影响。方法将人胃低分化粘液腺癌MGC-803细胞培养12小时后,分为对照组和薯蓣皂苷元(30μg·mL-1)干预24小时组。干预结束后,提取mRNA,干预组和对照组逆转录分别用Cy5和Cy3标记,成为两组cDNA探针。cDNA探针与基因芯片杂交,经扫描仪扫描,用软件分析Cy5和Cy3两种荧光信号的强度和比值。结果芯片结果显示筛选出差异表达基因共158条,其中101条表达下降,57条表达增加。结论通过基因芯片技术,发现多个基因的表达水平可受薯蓣皂苷元用药的影响,通过对细胞基因表达的深入研究,对进一步理解薯蓣皂苷元抗肿瘤的分子机制可能有帮助。To study the effects of Diosgenin on the expression of associated genes in human gastric cancer cells MGC-803 using cDNA microarray. Methods: Gastric cancer cells MGC-803 were incubated for 12h. All cells were into control group,exposure to diosgenin (30 ?g-mL-1 ) for 24h, and cDNA probes were prepared by labeling the mRNA with fluorescent Cy5 and Cy3 by reverse transcription in 2 groups, respectively. The arrays were hybridized against the cDNA probes and then the fluorescent signals were scanned. The intensity of fluorescent signal of Cy5 and Cy3 and the ratio of Cy5 to Cy3 were analyzed by software . Results: 158 gastric cancer-associated genes expressions were screened, and 101 of them were decreased and 57 of them were increased. Conclusion: There are evident effects of Diosgenin on the experimental gastric cancer-associated genes expressions in human It will be helpful for understanding the molecular mechanism of Diosgenin in gastric cancer.
