摘 要:目的 了解哈尔滨地区G1型A组轮状病毒VP7基因变异特点.方法 选取2007-2012年哈尔滨地区33株G1型A组轮状病毒标本,采用半巢氏PCR进行G1型鉴定,并对VP7基因进行克隆测序.采用DNASTAR软件对序列进行分析及进化树的绘制.结果 33株哈尔滨地方株VP7基因均为1062个碱基,推导氨基酸序列同源性95.7%~100%;33株哈尔滨地方株根据进化树分为四簇,其中Ⅰ簇17株,Ⅱ簇12株,Ⅲ簇、Ⅳ簇各2株.在氨基酸水平上,Ⅰ簇与其他三簇的整体差异位点为aa75(V→I),Ⅱ簇哈尔滨地方株变异比较活跃,Ⅱ簇、Ⅲ簇哈尔滨地方株具有一定的中国地方性,Ⅳ簇参考株均来自东南亚地区;2007年的哈尔滨地方株VP7蛋白变异位点最多,2008年次之,2010年-2012年哈尔滨地方株主要集中在Ⅰ簇.结论 33株轮状病毒G1型哈尔滨地方株可分为四个分支,Ⅰ簇是优势分支,且随着监测年份的变化Ⅰ簇所占构成比逐渐增加.Objective In order to understand the diversity of the VP7 genes among group A rota- virus G1 type strains in Harbin. Methods Thirty-three rotavirus strains of G1 type were selected in Harbin during 2007- 2012. G1 type was identified using the semi-nested PCR, and the VP7 genes were cloned and sequenced. The phylogenetic tree was constructed by DNASTAR software. Results The VP7 genes of all 33 strains were 1062 bp in size, and the amino acid identity was 95.7% - 100% among themselves. Phylo- genetic analysis revealed that 33 Harbin strains were divided into four clusters, 17 strains belonged to cluster I , 12 of cluster II , 2 of cluster III , and 2 of iV. At the amino acid level, compared with the other three clusters, mutations of aa75 (V→d) were found in all strains of Cluster I . More mutated sites were detected in Cluster II , and clusters II and III were often seen in our country. Cluster iV were highly homologous with the reference strains circulating in southeast Asia. Most mutated sites of VP7 protein among Harbin local strains were found in 2007, followed by 2008, the Harbin local strains mainly belonged to cluster I during 2010 to 2012. Conclusions Thirty-three Harbin strains of rotavirus G1 type were divided into four clusters by phylogenetic analysis, among which cluster I was predominant, and the proportion of cluster I was increased year by year.
