摘  要:目的:建立中药菟丝子中金丝桃苷、紫云英苷、槲皮素、山柰酚和异鼠李素的含量的高效液相色谱测定方法。方法:采用AgilentTCC18色谱柱(4.6mm×250mm,5μm),流动相为甲醇-0.4%磷酸,紫外检测器:检测波长360nm,流速为1mL/min,梯度洗脱:0→60min→80min,甲醇的比例由25%→40%→60%,柱温45℃,测定中药菟丝子中5种黄酮类成分的含量。结果:色谱峰分离情况良好,菟丝子中5种化学成分在各自的浓度范围内均具有良好的线性关系;加样回收率为98.0%-102.2%,RSD为1.44%-2.63%。结论:所建方法简便、准确,重现性好,可用于菟丝子药材的质量评价。Objective: To determine the content of five kinds of flavonoids in the seed of semen cuscutae, including hyperoside, astraga- lin, quercetin, kaempferol and isorhamnetin. Methods: The HPLC method was used to determine the content of flavonoids in the seeds of semen cuseutae. The analysis was carried out on Agilent TC C18 column (250 mm ×4. 6 mm, 5 μm), with MeOH-0. 4 % H3PO4as mobile phase, the flow rate of 1 mL · min-l and wave length at 360 nm. The gradient elute time was 0→min→80min, and the proportion of methanol was 25%→40%→60%. at45℃. Results: Chromatographic peak separation was in good condition, and five kinds of flavonoids had good linear relationship respectively within their own scope of concentration ; the recovery was 98.0% ~ 98. 0%, RSD was 1.44% ~ 2.63%. Conclusion:The established method is simple, accurate, and reproducible and can be used as the quality evaluation of semen cuscutae.
