摘 要:本研究主要是为了探讨体外受精过程中不同获能液及不同处理方法对精子获能的影响。本实验采用BO液和TCM199液2种获能液,分别使用上游法和洗涤法对精子进行获能处理,通过分析各组的精子存活时间、活精子百分率和顶体反应率,从而选择最佳的精子获能组合方法。结果表明:采用BO液和TCM199液2种获能液,分别使用上游法和洗涤法对精子获能处理,其对精子存活时间的影响差异均不显著(P>0.05);在相同获能液的条件下,2种不同的精子获能方法对活精子百分率的影响差异均不显著(P>0.05);在相同的精子获能处理方法条件下,采用BO液对活精子百分率的影响均显著高于TCM199液培养组(P<0.05);采用BO液培养,2种不同的精子获能方法对精子顶体反应率的影响差异显著(P<0.05),采用上游法处理时,2种不同获能培养液对精子顶体反应率的影响差异显著(P<0.05)。This study was conducted to investigate the effects of different culture mediums and different treatment methods on sperm capacitation. In this experiment, the BO medium and TCM199 medium were used as capacitation mediums by using swim-up method and direct washing method respectively to handle the semen. In order to choose the best method of sperm capacitation, the sperm survival time, the percentage of live sperm and acrosome reaction were detected. The results showed that: Neither the two different culture medium nor the two different treatment methods can lead to significant difference of the sperm survival time (P〉0.05); At the same culture medium, there was no significant difference of the two different processing methods on the percentage of live sperm (P^0.05), and at the same processing method, the effect of the BO medium on the percentage of live sperm was significant higher than the TCM199 medium (P〈0.05); While using the BO medium as capacitation medium, the two different processing methods had significant effects on the percentage of acrosome reaction (P〈0.05); while using the swim-up method, the two different culture mediums had highly significant effects on the percentage of acrosome reaction(P〈0.05).
