摘 要:目的探讨干扰素-γ(IFN-γ)及肺细胞DNA损伤在铍病发病中的作用。方法无特定病原体级健康SD雄性大鼠66只随机分为模型组、溶剂对照组和空白对照组。模型组采用一次性非暴露式气管内注入质量浓度为10g/L的氧化铍溶液0.5 ml,制备肺损伤模型;溶剂对照组采用相同方法注入等体积灭菌生理氯化钠溶液;空白对照组不给予任何处理。分别于模型制备后20、40、60 d,采用光学显微镜和透射电子显微镜观察肺组织损伤情况;酶联免疫吸附法检测肺组织匀浆和肺泡灌洗液中IFN-γ相对表达水平,单细胞凝胶电泳法检测肺细胞DNA链损伤情况。结果模型组大鼠肺组织病理学改变基本符合人类铍病特点。模型组20、40、60 d时间点大鼠肺组织匀浆和肺泡灌洗液中IFN-γ相对表达水平均分别高于相应时间点溶剂对照组和空白对照组(P<0.01);模型组60 d时间点大鼠肺组织匀浆和肺泡灌洗液中IFN-γ相对表达水平均分别高于20 d时间点水平[(100.51±24.37)vs(72.54±14.91),(119.51±25.42)vs(86.76±17.75),P<0.05]。模型组3个时间点肺细胞尾长、尾部DNA百分率和OLive尾距均高于相应时间点溶剂对照组和空白对照组(P<0.01);模型组40、60 d时间点肺细胞的尾长、尾部DNA百分率和OLive尾距均分别高于20 d时间点相应指标值(P<0.01)。结论氧化铍可引起大鼠肺组织匀浆及支气管肺泡灌洗液中IFN-γ相对表达水平的升高及肺细胞DNA的损伤,可能是导致铍病肺损伤的机制。Objective To explore the role of interferon ( IFN) -γand DNA damage of lung cells in the pathogenesis of experimental berylliosis in rats .Methods Sixty-six specific pathogen free healthy male SD rats were randomly divided into model group, solvent-and blank-control group.The model group was intratracheally injected with 10 g/L beryllium oxide 0.5 ml for preparing the lung injury model;solvent control group was injected with the same volume of sterilized normal solu -tion by the same method; blank control group was not given any treatment .Optical microscopy and transmission electron microscopy were used to observe the injury in lung tissue at day 20, 40 and 60 of modeling;enzyme-linked immunosorbent assay was used to detect the relative expression levels of IFN-γin lung tissue and bronchoalveolar lavage fluid ( BALF );single cell gel electrophoresis assay was used to detect the DNA strand damage in lung cells .Results The results showed that the pathological changes were consistent with the basic features of human berylliosis .IFN-γrelative expression levels were relatively higher in lung tissues and BALF in the model groups respectively (P&lt;0.01), compared with those of solvent-and the blank-control groups at the same time points of day 20, 40 and 60.In model group, IFN-γrelative expression levels in lung tissues and BALF on day 60 of modeling were significantly higher than those on day 20 [ ( 100.51 &#177;24.37 ) vs (72.54 &#177;14.91), (119.51 &#177;25.42) vs (86.76 &#177;17.75), P&lt;0.05].Lung cell tail length, tail DNA percentage and OLive tail index values in the model groups were higher than those in solvent-and the blank-control groups at the three corresponding time points respectively (P&lt;0.01).In the model groups, tail length, tail DNA percentage and OLive tail in lung cells on time points of day 40, 60 were higher than those on day 20 respectively (P&lt;0.01).Conclusion Beryllium oxide can induce IFN-γlevels increase and lung cell DNA dama
