摘  要:目的观察通心络超微粉对急性脑梗死大鼠脑微血管的影响,探讨其脑保护的作用机制。方法制作大鼠左侧大脑半球永久性大脑中动脉栓塞模型,随机分为假手术组、模型组、通心络组和丁苯酞组,每组再分为1、3 d 2个亚组。假手术组和模型组给予生理盐水灌胃,通心络组给予通心络超微粉灌胃,丁苯酞组给予丁苯酞灌胃。采用2,3,5-氯化三苯四唑染色检测大鼠脑组织梗死体积百分比,HE染色观察大鼠脑微血管的一般形态,免疫组化染色测定各组脑组织中血管生成素-1(ANG-1)、内皮抑素(ES)表达水平。结果与模型组比较,通心络组和丁苯酞组的脑梗死体积百分比均减小,脑梗死灶面积明显减小,水肿程度减轻;与模型组比较,通心络组和丁苯酞组1、3 d测定ANG-1表达均升高(P<0.05),且通心络3 d组明显优于其余各组(P<0.001);与模型组比较,通心络1 d组ES表达有升高趋势,丁苯酞1 d组明显升高(P<0.05),丁苯酞3 d组有降低趋势,通心络3 d组明显降低(P<0.05)。结论通心络超微粉能够减轻脑微血管的损伤程度,通过调节ANG-1和ES的表达,促进后期微血管的新生,起到脑保护作用。Objective To observe the effect of Tongxinluo superfine powder on angiogenesis in acute cerebral infarction in rats and explore the mechanism of cerebral protection. Methods With suture method, a side of the middle cerebral artery of rats was blocked to establish the permanent middle cerebral artery occlusion. After filtration, male SD rats were randomly divided into sham-operation group, model group, Tongxinluo group and Butylphthalide group, which were then divided into 2 subgrouops, namely, the 1st day group and the 3rd day group, and were given corresponding drugs. The cerebral infarction volume fraction was determined by TTC staining. The morphological structure changes of cerebral microvessels were observed by HE staining, and immune-hitochemical method was used to detect ANG-1 and ES changes. Results Compared with the model group, the cerebral infarction volume fraction of Tongxinluo group and Butylphthalide group were reduced and infarction size were significantly reduced, the water content of brain tissue dropped. Compared with the model group, ANG-1 expression of Tongxinluo group and Butylphthalide group increased at the first and third day (P〈0.05), while the 3rd day Tongxinluo group was the most stongly increased (P〈0.001). Compared with the model group, ES expression of the 1st day Tongxinluo group tended to increase, the 3rd day Butylphthalide group was significantly higher (P〈0.05), the 3rd day Butylphthalide group tended to decrease, the 3rd day Tongxinluo group was significantly lower (P〈0.05). Conclusion Tongxinluo superfine powder can reduce the extent of cerebral microvessels lesions. Tongxinluo shows obvious protective effects on angiogenesis by regulating the expression of ANG-1 and ES.
