大豆GAMYB基因序列分析及拟南芥同源突变体鉴定

摘  要:GAMYB基因是赤霉素(GA)信号转导途径中的一个重要促进因子,该基因与植物由营养生长向生殖生长的开花过渡以及生殖发育有重要关系。通过构建系统进化树分析,发现该基因与拟南芥MYB33和MYB65亲缘关系较近,根据TAIR网站上公布的GAMYB-Like基因信息,从美国拟南芥生物资源中心购买该基因的T-DNA插入拟南芥突变体材料,运用双引物法对此突变体进行T-DNA插入位点的鉴定,分别获得3株和6株稳定遗传的纯合突变体,可为深入研究GmGAMYB基因的功能及进行互补拟南芥突变体myb33或myb65试验奠定基础。GAMYB gene is an importantly promotive factor in the signal transduction pathways of gibberellin (GA).The gene played an important role in plant flowering transition from vegetative growth to reproductive growth and reproductive development. In order to determin wether a similar biological function existed between GmGAMYB gene of Glycine max and the one of Arabidopsis a phylogenetic tree was constructed according to GAMYB sequence data. GmGAMYB gene was closely related to Arabidopsis MYB33 and MYB65. According to the information published on TAIR website, the homozygous genetic T-DNA insertion mutant of GAMYB-Like gene were obtained from TAIR. Using double primer method identified this T-DNA insertion mutant, respectively obtained three and six stably inherited homozygous mutant.These homozygous mutant lay a good foundation for further investigating the function of GmGAMYB gene in complementing Arabidopsis myb33 or myb65 mutants.
