摘  要:SND1(p100)作为转录共激活子,在信号转导和转录激活因子5(STAT5)中起重要作用,已在牛以外的其他物种中报道SND1与STAT5相互作用激活STAT5基因介导基因转录。试验为验证SND1对奶牛乳腺上皮细胞增殖及细胞周期影响,构建SND1表达载体及si RNA转染奶牛乳腺上皮细胞,用CASY法检测细胞活力及增殖能力,用流式细胞仪检测奶牛乳腺上皮细胞周期,Western Blot确定SND1转染奶牛乳腺上皮细胞功能。结果表明,SND1对于奶牛乳腺上皮细胞增殖属于正调控因子,SND1与STAT5相互作用影响CyclinD1表达,SND1通过CyclinD1途径促进细胞周期进行。SND1 (p100) as a co-activator for several transcription factors, plays a vital role in signal transduction and activator of transcription 5(STAT5), there are reports that SND1 interaction with STAT5 mediates gene transcription. In this study, in order to verify SND1 effects on cell proliferation and cycle distribution of dairy cow mammary epithelial cells (DCMEC), we constructed transfected DCMEC with SND1, the viability and proliferation of DCMEC were measured by CASY method, DCMEC were detected by flow cytometry method. Western blot was used to determine SND1 transfection into DCMEC function. These data demonstrated that SND1 was a positive regulatory factor of DCMEC on proliferation ability, and SND1 interacted with STAT5 which impacted CyclinD1 expression. SND1 could promote cell cycle by CyclinD1 pathway.
