摘 要:目的观察不同药物对鱼胆毒素(鲤醇硫酸酯钠)致人肾近曲小管上皮细胞(HK-2)损伤的影响。方法从新鲜草鱼胆汁中提取鲤醇硫酸酯钠,通过体外培养HK-2细胞系,应用MTT法评价鲤醇硫酸酯钠对HK-2细胞增殖抑制作用,并观察丹参、地塞米松、氨氯地平、水飞蓟宾、腺嘌呤核苷三磷酸(ATP)多种药物对鲤醇硫酸酯钠染毒HK-2细胞的干预效果。结果 (1)HK-2细胞存活率随着鲤醇硫酸酯钠浓度的增加和时间的延长而降低;(2)丹参可促进HK-2细胞增殖,HK-2细胞经丹参预处理再鲤醇硫酸酯钠染毒24h后,细胞存活率显著增加,并随丹参浓度的增加而增加(P<0.01),但地塞米松、氨氯地平、水飞蓟宾、腺嘌呤核苷三磷酸(ATP)干预鲤醇硫酸酯钠染毒HK-2细胞后,其细胞存活率无明显增加。结论丹参可有效降低鱼胆汁毒素致肾小管上皮细胞的损伤,提高细胞活力。Objective To observe the effect of different drugs on human renal epithelial cells inju- ry induced by ichthyogalltoxin (sodium cyprinol sulphate, SCS). Methods SCS was extracted from the grass carp bile according to method of the literature reported. Human proximal tubular epithelial cell line (HK-2) was cultured in vitro. Methyl thiazolyl tetrazolium (MTT) assay was used to evaluate the multiplication-inhibiting of SCS on HK-2 cells, and the effects of different drugs (salvia miltiorrhiza, dexamethasone, amlodipine, silybin, and adenosine triphosphate ) on the viability of HK-2 cells trea- ted by SCS. Results The viability of HK-2 cells induced by SCS was significantly reduced with concen- tration and time dependant. The salvia miltiorrhiza could promote HK-2 cells multiplication. After HK-2 cells were pre-treated with salvia miltiorrhiza and contaminated by SCS for 24 hours, HK-2 ceils' viability was significantly increased along with the salvia miltiorrhiza concentrations increasing (P〈0.01), while this feature did not demonstrated in other drugs including of dexamethasone, sily- bin, amlodipine, and adenosine triphosphate. Conclusion The salvia miltiorrhiza can effectively prevent HK-2 cells from damage caused by SCS and improve the HK-2 cells viability.
