摘  要:以甜叶菊茎段为外植体,采用不同低浓度秋水仙素在甜叶菊试管苗茎段再生苗初期进行长时间持续处理,以探讨低浓度秋水仙素诱导甜叶菊多倍体的优势及适合长时间诱导甜叶菊的秋水仙素浓度,简化甜叶菊离体诱导多倍体技术环节。结果表明:0.025%的秋水仙素浓度适合于甜叶菊多倍体离体诱导的持续诱变,直至试管苗再生而无需清洗外植体和更换培养基,简化了诱导技术环节,提高了甜叶菊多倍体离体诱导效率;多倍体植株田间农艺性状表现为叶片大而厚实、开花延迟、花蕾较大等特征。旨在建立低浓度秋水仙素离体诱导甜叶菊多倍体技术体系。The stem sections of S. rebaudiana Bertoni were used as explants for polyploidy induction in vitro by low concentrations of colchicines for long time treatments during shoot formation. The aims were to study the superiority of polyploidy induction by lower concentrations of colchicines for long time in S. rebaudiana Bertoni and simplify the technique protocols for polyploidy induction in vitro in S. rebaudiana Bertoni. The results showed that the concentration of 0.025% colchicines was suitable to induct polyploidy in vitro for long time treatment and need not rinse explants, change medium until the shoots formation. Agricultural traits of the polyploidy displayed bigger and thicker leaves, bigger flower bud and florescence being postponed.
