参芪益心方对CHF大鼠心肌细胞凋亡指数及Bax、Bcl-2表达的影响

摘　　要：目的:研究参芪益心方对阿霉素所致慢性心力衰竭(CHF)大鼠心肌细胞凋亡指数及Bax、Bcl-2表达的影响及其作用机制。方法:将90只雄性大鼠随机平均分为6组:正常对照组、模型组、依那普利对照组、芪苈强心胶囊对照组、参芪益心方低剂量组、参芪益心方高剂量组。利用阿霉素复制CHF大鼠模型,Tunel法检测心肌细胞凋亡指数,免疫组化法测定心肌细胞凋亡基因Bax、Bcl-2的表达。结果:各治疗组CHF大鼠心肌凋亡指数均有所降低,且与模型组比较具有统计学意义(P<0.05)。各治疗组均能抑制CHF大鼠心肌细胞Bax基因的表达;同时各治疗组能够促进大鼠心肌细胞Bcl-2基因的表达,且与模型组比较具有显著性差异(P<0.05),其中以参芪益心方高剂量组为优。结论:参芪益心方可能调控凋亡基因从而抑制心肌细胞凋亡,进而延缓心室重塑的发生、发展或部分逆转心室重塑,从而起到治疗CHF的效果。Objective：To study the effect of Shenqi Yixin formula on myocardial apoptosis index,expression of Bax and Bcl-2 in chronic heart failure rats induced with doxorubicin. Methods：Ninety male rats were randomly divided into six groups： normal control group,model group,enalapril control group,the Qili Qiangxin capsule control group,the low dose Shenqi Yixin formula group,and the high dose Shenqi Yixin formula group. Chronic heart failure rat models were established using doxorubicin. Assay apoptosis rate via terminal deoxynucleotidyl transferase dUTP nick end labeling（TUNEL） essay was applied to test the myocardial apoptosis index,and expressions of Bax and Bcl-2 expression were tested with immunohistochemical methods. Results：Myocardial apoptosis indices of chronic heart failure rats in treatment groups were decreased with statistical difference compared with the model group（P〈0.05）. The expression of Bax in each treatment group cardiomyocytes was decreased while the expression of Bcl-2 gene was increased,with statistical differences compared with the model group（P〈0.05）the effect in high dose Shenqi Yixin formula group was the most obvious. Conclusion：Shenqi Yixin formula could modulate apoptotic genes to inhibit apoptosis, and therefore it could slow or reverse ventricular remodeling. Thus it could treat the chronic heart failure.
