摘 要:目的:应用异硫氰酸荧光素(FITC)诱导的Th2型小鼠变应性接触性皮炎的模型,评价黄芪甲苷(AS-IV)对Th2过敏性皮炎的作用。方法:BALB/c小鼠在day 1,day 2用1.5%的FITC溶液致敏腹部皮肤2次,同时给予地塞米松ip剂量为0.67 mg·kg-1,AS-IV(6.25,12.5,25,50 mg·kg-1,ig)至day 5(诱导相给药),day 6用0.5%FITC溶液攻击小鼠耳部皮肤,24 h后测定小鼠耳肿胀程度,耳组织HE染色,同时部分耳组织制匀浆以ELISA方法检测白介素(IL)-4,IL-13,IL-9水平。结果:与正常组比较,模型组小鼠耳肿胀明显,耳组织真皮层明显水肿,炎症细胞大量浸润,Th2型细胞因子IL-4,IL-13,IL-9水平显著升高;黄芪甲苷25,50 mg·kg-1诱导相给药能显著抑制模型的耳肿胀度;病理组织学检查亦显示与模型组比较,给予黄芪甲苷小鼠的耳炎症反应减轻,炎细胞浸润明显减少;同时能显著降低耳组织匀浆中IL-4,IL-13,IL-9水平。结论:黄芪甲苷诱导相给药对FITC诱导的小鼠Th2型变应性接触性皮炎有显著的抑制作用,该作用与抑制Th2型细胞因子的产生有关。Objective: Mice Type2 allergic contact dermatitis induced by fluoresciniso thiocyate (FITC) was utilized in this study to explore the effect of astragaioside IV (AS-IV) on allergic inflammatory. Method : The BALB/c mice were topically sensitized on the shaved abdomens with 1.5% FITC solution at day 1 and day 2 and elicited on the right ear with 0.5% FITC at day 6. Mice in the dexamethasone group were intraperitoneal injection with dexamethasone (0.67 mg·kg^-1) and the AS-IV group were intragastrically administered with astragaioside IV (6.25, 12.5, 25, 50 mg ·kg^-1) from day 1 to day 5 (induction phase). Auricle swelling was measured 24 h after the elicitation. Pathohistological examination was carried out by HE staining. Interleukin-4 ( IL-4), IL-13 and IL-9 levels of ear tissue homogenates were detected by ELISA. Result: Compared with the normal group, ear swelling in model group was stimulated by FITC treatment significantly and ear tissue local edema, angiectasis and lymphocytic infiltration were observed simultaneously. Meanwhile, IL-4, IL-13 and IL-9 levels increased remarkably in FITC treated mice. Wherein, AS-IV 25 mg·kg^-1 and 50 mg·kg^-1 evidently inhibited the auricle inflammation of the FITC treated mice. Pathohistological results indicated that AS-IV alleviated local edema and angiectasis of mice models and reduced lymphocytic infiltration significantly. Furthermore, IL-4, IL-13 and IL-9 levels in ear tissue homogenates were markedly decreased by AS-IV treatment. Conclusion: Administration with AS-IV in induction phase could inhibit Th2 cell-mediated allergic contact dermatitis in mice significantly and this effect was related with regulation on type 2 cytokines.
