摘  要:目的观察慢性应激抑郁模型大鼠主动脉AngⅡ、AT1R、ACEⅠ表达的影响以及电针的干预作用,探讨肾素-血管紧张素系统(RAS)在抑郁病与心血管疾病之间的关联。方法采用慢性应激刺激结合孤养的方式建立大鼠慢性应激抑郁模型,分组后分别采用电针、氟西汀治疗21d,通过敞箱试验观察大鼠的行为学变化;免疫组化法(S-P)测定主动脉AngⅡ、AT1R、ACEⅠ表达。结果模型组大鼠敞箱试验水平运动得分和垂直运动得分明显降低,电针和氟西汀可逆转此变化;模型组大鼠主动脉组织AngⅠ、ACE、AT1R高度表达,电针和氟西汀均可降低变化。结论慢性应激抑郁模型大鼠的局部RAS被过度激活,电针和氟西汀可降低其激活程度。Objective To observe the changes of angiotensinⅡ(AngⅡ),angiotensinⅡtype 1(AT1R)and angiotensin converting enzyme 1(ACEⅠ)in aorta tissue with chronicity stress depression model rats and the interference of electro-acupuncture to them,and to explore the function of the renin-angiotensin system(RAS)between depression and cardiovascular disease.Methods The chronic unpredictable mild stress combined with solitarily feeding method was used to make the depression model.The locomotor activity of rats in blank control group,model control group,electro-acupuncture group and fluoxetine group was observed by open-field experiment.The expression of AngⅡ,AT1Rand ACEⅠ were analyzed by immunohistochemistry method.Results The cross and rear score of the model group are significantly lower than blank group,while electro-acupuncture and fluoxetine reverse it.The expression of AngⅡ,AT1Rand ACEⅠin the arterial tissue of model rats enhanced.Electro-acupuncture and fluoxetine Couaol improve those pathological changes.Conclusion Local tissue RAS was activated excessively in chronicity stress depression model rats,and electroacupuncture and fluoxetine Couel improve those pathological changes.
