Effects of protein-free calf blood extract and recombinant human epithelial growth factor on mechanical corneal epithelial wound healing and neovascularization of rabbits

摘  要:目的探讨小牛血去蛋白提取物与重组人表皮生长因子(recombinant human epithelial growth factor,rhEGF)对兔角膜上皮损伤修复及血管新生的作用。方法建立兔角膜上皮损伤及新生血管模型,随机分为三组(每组5只5眼):小牛血去蛋白提取物组(速高捷组)、rhEGF组和生理盐水组(NS组)。术后1 d开始分别滴速高捷、rhEGF和生理盐水,每天4次,观察角膜上皮损伤修复及新生血管面积变化。结果术后1 d,rhEGF组、速高捷组与NS组角膜上皮愈合率分别为(51.83±8.87)%、(39.35±6.92)%、(43.32±5.42)%,三组间差异有统计学意义(F=6.475,P=0.012),rhEGF组角膜上皮愈合率高于速高捷组(P=0.004)和NS组(P=0.041)。术后2 d、3 d,三组间角膜上皮愈合率差异均无统计学意义(均为P>0.05)。术后18 d,速高捷组角膜新生血管面积(58.27±5.42)mm2小于rhEGF组(79.98±8.63)mm2和NS组(77.16±12.59)mm2,差异均有统计学意义(P=0.040、0.008)。术后8 d、12 d、15 d和21 d,三组间角膜新生血管面积差异均无统计学意义(均为P>0.05)。结论 rhEGF早期(2 d内)较速高捷促进兔角膜上皮损伤修复作用明显,rhEGF和速高捷促进角膜新生血管作用不明显。Objective To investigate the effects of recombinant human epidermal growth factor (rhEGF) and protein-free calf blood extract (PCBE) on corneal epithelial wound healing and corneal neovascularization (CNV) of rabbits. Methods The corneal wound and CNV model was established in rabbits, and randomly divided into three groups :PCBE group,rhEGF group and NS group,5 cases (5 eyes) in each group. One eye of each group was chosen randomly for topical administration of PCBE, rhEGF or NS, and variability in the area of corneal epithelial wound healing and CNV were observed. Re- suits On day 1, the healing rates of corneal epithelium in rhEGF group, PCBE group and NS group were (51.83 ± 8.87 ) % , (39.35 ± 6.92) % and (43.32 ±5.42) % , respective- ly, there was statistical difference among three groups (F = 5.475 ,P = 0.012). The healing rate of corneal epithelium in rhEGF group was higher than those in PCBE group and NS group ,there were statistical differences (P =0.004,0. 041 ). On day 3 ,there was no statis- tical difference in the healing rate of corneal epithelium among three groups (all P 〉 0.05). On day 18,the area of CNV in PCBE group, rhEGF group and NS group were (58. 27 ±5.42)mm2 ,(79.98 ±8.63)mm2 and (77.16±12.59)mm2 ,respectively,which in PCBE group was less than those in rhEGF group and NS group, there were statistical differences (P =0. 040,0.008). There was no statistical difference in CNV area on day 8, 12,15,21 among three groups ( all P 〉 0.05 ). Conclusion rhEGF has better promotive effect on corneal epithelial wound healing than PCBE in the advanced phase (within 2 days). Both rhEGF and PCBE has no obviously effect on CNV.
