摘 要:目的优选救必应提取物的制备工艺。方法采用单因素法选择提取溶剂;采用正交试验法,以长梗冬青苷质量分数、浸膏得率为指标,考察提取次数、提取时间、提取溶剂用量3个因素对提取工艺的影响;采用反相高效液相色谱法测定长梗冬青苷质量分数,烘干法测定浸膏得率。以长梗冬青苷质量分数为指标优选减压浓缩和干燥工艺。结果救必应最佳提取工艺为:药材加8倍量70%(体积分数)乙醇,加热回流提取2次,每次1 h;在60℃下减压浓缩和干燥。结论该工艺设计简便、稳定可行,适用于工业化生产。Objective To optimize the preparation process of the extract from Ilex rotunda Thunb.. Methods One-way analysis was used to select the solvent. Orthogonal design was designed to investigate the amount of solvent,extracting times and time with the content of pedunculoside and yield of extract as indexes.The content of pedunculoside was assayed by RP-HPLC, and the yield of extract was determined by oven drying method.The content of pedunculoside was used to investigate the process of drying and concentrating under reduced pressure. Results The best extraction conditions were as follows: refluxing twice with eight-fold 70% ethanol, for one hour each time. The temperature of drying and concentrating under reduced pressure was 60 ℃. Conclusion The optimized extraction process was reasonable, stable, feasible, which may be applicable to industrial production.
