摘 要:目的探索5型病毒载体的保存条件,使病毒载体在保存过程的活性损失最小,对类癌细胞的杀伤作用最大。方法采用正交设计法,考虑温度、甘油含量及MgCl2浓度3因素,对病毒载体采用了3因素3水平的试验,测出不同保存配方的病毒载体的滴度。同时检测病毒载体在体对类癌细胞杀伤的影响。结果在-80℃,含5%甘油和3 mmol/L MgCl2的保存条件下病毒载体的活性最高,对类癌细胞HEK-293杀伤作用最明显;温度对病毒载体的活性影响最显著,甘油含量及MgCl2浓度对病毒载体的活性影响并不显著。结论温度越低,病毒载体体内反应越低;滴度越大,对类癌细胞的杀伤效果越大。温度越低对病毒载体的保存越有利,同时也为其他病毒载体保存条件的研究提供借鉴。Objective To explore the preservation conditions of adenovirus type 5 for reduction of adenovirus 6ters loss and maximization of the cytotoxic effect on cancer cells. Methods Orthogonal design method was used, and three factors, namely the temperature, glycerol content and concentration of MgCl2 were considered.Three factors and three levels of tests were applied to measure different preservation formulations of adenovirus titers.Meantime, the effect of adenovirus on killing cancer cells was detected.Results The best condition for adenovirus preservation included -80 ℃, 5% glycerol and 3 mmol/L MgCl2, and maximized the cytotoxic effect on cancer cells.The most significant factor was the temperature, the effect of glycerol and MgCl2 content was not as important as the temperature.Conclusion The lower temperature is beneficial for adenovirus preservation, and the eytotoxic effect on cancer cells is increased as higher titer of adenovirus. This study also provides a reference for other adenovirus preservation.
