摘 要:近年来,已报道从成年的小鼠中分离得到心脏干细胞(Cardiac Stem.Cells CSCs),并且应用于心脏病的治疗.但是心脏干细胞特征尚未完全阐述.通过体外分离成年小鼠的心肌组织,进行培养,分离得到半悬浮状态的具有干细胞特性的细胞;利用流式细胞仪检测分离得到的心脏干细胞的表面分子表达情况;并且通过调整培养条件诱导其体外分化.研究结果显示成功分离得到的心脏干细胞,表达干细胞标志分子Sca-1,以及间充质干细胞标志分子(CD29、CD90、CD44和CD106);体外分化研究显示心脏干细胞可以分化成心肌细胞、平滑肌细胞、脂肪细胞和成骨细胞.心脏干细胞具有间充质干细胞的特性,具有多向分化潜能,在心脏损伤的治疗中具有重要意义.Cardiac Stem Ceils (CSCs) have been isolated from heart of adult mouse and hu- man, and have shown beneficial effects in the therapy of ischemia heart disease. However, the characters of the CSCs were not fully elucidated. In this study, myocardial tissues from adult mouse heart were minced, digested and cultured. The phase-bright cells were harvested after subculture. In vitro characterization was done by fluorescence-activated cell sorting (FACS) analysis. In vitro differentiation of CSCs was carried out. The results demonstrated CSCs were obtained successfully; FACS analysis data revealed that this population cells express resident CSC markers (Sca-1) and exhibit mesenchymal features with expression of mesenchymal stem cell markers (CD29, CD90, CD44 and CD106). Afterwards, these cells can differentiate into cardiomyocytes, smooth muscle ceils, adipocytes and osteoblasts. In conclusion, CSCs exhibit mesenchymal features and can successfully differentiate into different cell lineages.
