摘  要:建立了HPLC-DAD法同时测定木蝴蝶中4种化学成分的含量。采用Agilent Eclipse XDB-C18色谱柱(4.6 mm×250 mm,5μm),以乙腈-0.1%冰醋酸水溶液进行梯度洗脱,流速:1 mL/min,柱温:30℃,检测波长:320 nm,进样量10μL。结果表明:芦丁、木犀草苷、槲皮素、黄芩苷4个化合物在测定的范围内均呈现出良好的线性关系(r≥0.9995),回收率为94.0%~98.0%。该方法操作简便,分离效果较好,灵敏度高,重现性良好,为较全面控制木蝴蝶药材的质量提供了一种可靠的参考方法。A novel HPLC analysis method coupled with DAD detector was developed and validated for the simultaneous determination of the contents of Rutin, Luteoloside, Quercetin and Baicalin in Oroxylum indicum. Chromatographic separation was successfully conducted by using Aglient Eclipse XDB - C18 column (4.6 mm ÷ 250 mm id, 5 μm) , eluted with a gradient mobile phase of acetonitrile and water -0. 1% glacial acetic acid at a flow rate of 1.00 mL/min, and detected at 320 nm. The temperature and the injection volume were set at 30 ℃ and 10 μL. Parameters of linearity, precision, accuracy and limits of detection and quantitation were considered. Within the range of the measurement, these four components presented a good linear relationship (r≥0. 9995) and the recovery was 94. 00% -98.00%. The validated method succeeded in determining the contents of four chemical components. This method had the advantages of simple operation, good separation, high sensitivity and good reproducibility, and also provided a reliable reference method for comprehensively controlling the quality of the Oroxylum indicum.
