摘 要:目的探讨应用养血清脑颗粒对血管性痴呆大鼠认知功能的影响,并观察大鼠海马细胞凋亡及p38MAPK磷酸化表达的变化。方法应用两血管法(2VO)制作血管性痴呆大鼠模型,将60只3月龄雄性Wistar大鼠随机分成血管性痴呆模型组、假手术组和养血清脑颗粒组。养血清脑颗粒组给予2 g·kg-1·d-1的养血清脑颗粒溶液2 ml灌胃,血管性痴呆组和假手术组给予等量的2 ml生理盐水灌胃,1个月后应用Morris水迷宫实验分别测试各组大鼠的空间认知能力,TUNEL法检测海马CA1区细胞凋亡,蛋白印迹法观察大鼠海马区p38MAPK磷酸化变化。结果第1、2、3天养血清脑颗粒组大鼠隐蔽平台逃避潜伏期明显小于模型组隐蔽平台逃避潜伏期,差异有统计学意义(P<0.01);养血清脑颗粒组大鼠原平台象限时间大于模型组,差异有统计学意义(P<0.01)。大鼠海马区细胞凋亡、磷酸化P38MAPK的表达的比较:养血清脑颗粒组明显低于血管性痴呆模型组,差异有统计学意义(P<0.01)。结论养血清脑颗粒能显著改善血管性痴呆大鼠的学习记忆能力,同时抑制海马区细胞凋亡,可能是通过抑制p38MAPK通路而实现的,这可能是养血清脑颗粒参与治疗血管性痴呆的作用机制之一。Objective To study the effects of Yangxue Qingnao Granule on cognitive function of rat model of vascular dementia, moreover observe the expression of apoptosis and p-p38MAPK in hippocampus. Methods In this research the vascular dementia (VD) model was established by two vascular (2VO) method, and then sixty male Wistar rats were randomly divided into VD group, sham operation group and YXQN (Yangxue Qingnao Granule) group. Rats in YXQN group were given 2 g·kg-1·d-1 dose Yangxue Qingnao Granule by gavage, moreover rats in VD group and sham operation group were given the same dose normal saline. The cognitive function of each rat was tested by Morris water maze, in addition the expression of p-p38MAPK in the hippocampus was observed by Western blot method;apoptosis was observed in hippocampal CAl region by TUNEL method. Results 1, 2, 3 day the hidden platform escape latency of YXQN group was significantly shortened (P〈0.01) compared with VD group;and the former platform quadrant time of YXQN group were significantly prolonged compared with VD group (P〈0.01); apoptosis and the expression of p38MAPK phosphorylation in hippocampus in YXQN group significantly reduced compared with VD group (P〈0.01). Conclusions Yangxue Qingnao Granule can significantly improve the learning and memory ability of rats model of vascular dementia, reduce apoptosis in the hippocampus by the inhibition of the p38MAPK pathway. This may be one of the ways by which Yangxue Qingnao Granule could treat vascular dementia.
