摘 要:目的:探讨补肾壮筋汤抑制炎性细胞因子表达延缓骨关节炎软骨退变的作用机制。方法:8周龄SD大鼠40只,随机分为正常组(10只)和造模组(30只)。造模组予质量分数4%木瓜蛋白酶复制骨关节炎模型后,随机分为模型对照组、补肾壮筋汤组、阳性对照组,每组10只。正常组常规喂养,模型对照组以生理盐水灌胃,补肾壮筋汤组以10.5 g·kg-1·d-1补肾壮筋汤灌胃,阳性对照组以150 mg·kg-1·d-1盐酸氨基葡萄糖胶囊溶液灌胃,每日1次,持续给药12周。采用HE、番红O-固绿染色观察软骨形态结构的变化,ELISA法检测关节液中基质金属蛋白酶-3、白细胞介素-1β、肿瘤坏死因子-α、透明质酸含量的变化。结果:软骨基质中蛋白多糖含量、胶原纤维的变性程度、软骨退变程度,补肾壮筋汤组与阳性对照组明显好于模型对照组。关节液中白细胞介素-1β、肿瘤坏死因子-α、基质金属蛋白酶-3的含量,补肾壮筋汤组与阳性对照组明显低于模型对照组(P<0.05);关节液中透明质酸的含量,补肾壮筋汤组与阳性对照组明显高于模型对照组(P<0.05)。结论:补肾壮筋汤通过抑制炎性细胞因子表达,延缓骨关节炎的软骨退变。Objective:To investigate the inhibitive effects of Bushen Zhuangjin Decoction on inflammatory cytokine expression to delay the degeneration of articular cartilage.Methods:40 SD rats of 8 weeks old were randomly divided into the normal group(10 rats) and the model group(30 rats).After replication of osteoarthritis models with 4%mass fraction of papain,the model group were randomly and equally divided into the model control group,the Bushen zhuangjin Decot ion group,and the positive control group.The normal group were routinely feed;the model control group were given lavage with normal saline;the BuShen Zhuangjin Decoction group were given Bushen Zhuangjin Decoction(10.5 g·kg-1·d-1), and the positive control group were given lavage with Glucosamine Hydrochloride Capsules solution(150 mg·kg-1·d-1). Al above treatments were done for 12 weeks.HE and safranin O-fast green staining were used to observe the changes of cartilage morphology.ELISA was used to detect the content of MMP-3,IL-1β,TNF-α and HA in synovial fluid. Results:The Bushen Zhuangjin Decoction group was better than the positive control group on the aspects of the contents of proteoglycans in the cartilage matrix,the denaturation of collagen ifber,and the degeneration of cartilage.The contents of IL-1β,TNF-α,and MMP-3 in synovia of the Bushen Zhuangjin Decoction group and the positive control group were signiifcantly lower than those in the model control group (P〈0.05). Conclusion:Bushen Zhuangjin Decoction can delay osteoarthritis cartilage degeneration by inhibiting the expression of inlfammatory cytokines.
