摘 要:目的观察抗行灵方的主要药用成分齐墩果酸和丹酚酸在单侧输尿管梗阻(UUO)模型大鼠肾间质纤维化(RIF)过程中的作用及机制。方法健康雄性SPF级SD大鼠48只,随机分为4组,其中假手术组、UUP模型组、齐墩果酸组、丹酚酸组各12只。造模后第28日腹主动脉采血检测血清肌酐(SCr)、尿素氮(BUN),收集24 h尿液,检测24 h尿蛋白定量、取肾组织,HE染色比较各组肾小管间质损伤程度。结果在UUO模型中,丹酚酸和齐墩果酸在延缓实验大鼠肾功能衰竭中的功能和侧重各有不同,除假手术外各组大鼠肾功能出现失代偿,尿微量蛋白排泄增多,肾间质纤维化加重;与假手术组、模型组相比,齐墩果酸组SCr、NAG、24 h尿蛋白定量的水平显著降低;丹芬酸组BUN、NAG、α1-M、24 h尿蛋白定量的水平显著降低(P<0.05或P<0.01)。结论丹酚酸及齐墩果酸单体均能够保护肾功能,降低血肌酐和尿素氮,并在一定程度上减少尿微量蛋白的排泄。Objective: To observe clinical effects of Kangxianling Decoction on kidney protection and to investi- gate function and mechanism of oleanolic acid and salvianolic acid on UUO rat model during renal interstitial fi- brosis. Methods: 48 healthy male SPF Sprague-Dawley rats were randomly divided into sham-operated group, model group,oleanolic acid group and salvianolic acid group, 12 in each group. Levels of serum creatinine (SCr), blood urea nitrogen (BUN),24 hpro from 24 h urine,HE staining,and nephridial tissue were collected and com- pared for the damage after 28 days. Results:In UUO model,oleanolic acid and salvianolic acid played different roles and functions in delaying renal failure. Except for sham-operated group,renal function of rats in other groups was deteriorated while excretion of urine micro protein gradually increased and renal interstitial fibrosis gradually deteriorated. Compared with sham-operated group and model group, the levels of SCr, NAG, and 24hpro reduced significantly while in oleanolic acid group the levels of SCr, NAG,24 hpro and α1-M lowered too (P 〈 0.05 或 P〈 0.01 ). Conclusions:Oleanolic acid and salvianolic acid can improve renal function and renal tubular function, reduce Scr and UN, and decrease the excretion of urine micro protein. But the mechanisms are different.
