电针小海与下巨虚穴对十二指肠溃疡大鼠血清肿瘤坏死因子-α及十二指肠核因子-κB的影响  被引量:12

Effects of Electroacupuncture on Serum TNF-α Level and Duodenal Tissue NF-κB Expression in DU Rats

摘  要:目的观察电针十二指肠溃疡(Duodenal ulcer,DU)大鼠"小海"、"下巨虚"后对血清肿瘤坏死因子-α(Tumour Necrosis Factor-α,TNF-α)含量及十二指肠组织核因子-κB(nuclear factor kappa-B,NF-κB)表达的影响。方法将40只健康SD大鼠随机分为空白、模型、小海、下巨虚四组,每组雌雄各半,除空白组外均皮下注射盐酸半胱胺建立DU模型大鼠。造模成功并治疗10 d后,肉眼和光镜下分别观察大鼠十二指肠黏膜溃疡情况及组织形态学变化,ELISA法检测大鼠血清中TNF-α含量,免疫组织化学法检测十二指肠NF-κB的表达。结果与空白组比较,各组大鼠光镜下十二指肠组织病理损伤改变明显,DU肉眼评分、血清TNF-α含量及十二指肠组织NF-κB表达显著增高(P<0.01);与模型组比较,小海、下巨虚组组织损伤有所改善,且DU肉眼评分、血清TNF-α含量及NF-κB表达显著降低(P<0.01);与小海组比较,下巨虚组组织损伤恢复较好,且肉眼评分、血清TNF-α含量及NF-κB表达降低(P<0.01或P<0.05)。结论电针小海、下巨虚穴均能对DU起治疗作用,且下巨虚组效果优于小海组,部分证实"合治内府"之"合"主要应指下合穴。其治疗效应可能是通过抑制核因子的活性、降低血清TNF-α等促炎因子的含量来实现的。Objective By electroacupuncture at Xiaohai and Xiajuxu points on duodenal ulcer model rats, to observe the effect of electroacupuncture on serum TNF-α level and duodenaltissue NF-κB expression, and to analyze their mechanism to which they related, briefly explaining the meaning of He points in curing viscera diseases by He points. Methods Forty SD rats were randomly divided into 4 groups: Blank group, Model group, Xiaohai group and Xiajuxu group. Set up the Duodenal Ulcer rat models through injection of cysteamine hydrochloride at the right buttock. After molding, following electro-acupuncatre at rat's XiaoHai and XiaJuXu acupoint, EA parameters density wave (10/50 Hz), a negative electrode on the right and the positive one left, the intensity between 1-3 mA, with limb tremor in degrees needle for 30 minutes once a day for 10 days. After treatment, make DU score by eyes first. Histological changes in the duodenum were observed by light microscopy. ELISA is used to check models' serum TNF-α level and the expression of NF-κB in the duodenum were detected by immunohistochemistry. Results The DU score, serum TNF-α levels and NF-κB expression of Model, Xiaohai and Xiajuxu groups are significantly higher than the Blank group (P〈0.01) and the tissue injurychanges are obviously under light microscope. Compared with Model group, the tissue injury changes of Xiaohai and Xiajuxu groups improved obviously, the DU score, serum TNF-α levels and NF-κB levels of Xiaohai and Xiajuxu groups are significantly lower (P〈0.01). Compared with Xiaohai group, the tissue injury changes of Xiajuxu group improved well, the DU score, serum TNF-α and NF-κB levels is obviously lower (P〈0.01 or P〈0.05). Conclusion Electroacupuncture at Xiaohai and Xiajuxu point have treatment effects on DU rats, and the effects of Xiajuxu is obvious better than Xiaohai, which proves Xiajuxu has relative specificity.
