摘 要:目的探讨急性心肌梗死心血瘀阻证模型大鼠心肌微环境p38信号转导通路对骨髓间充质干细胞(BMSCs)分化的影响。方法 48只SD大鼠随机分为健康对照组、假手术组(急性心肌梗死模型)、模型组(急性心肌梗死心血瘀阻证模型)、注射1组(急性心肌梗死心血瘀阻证模型大鼠于梗死区边缘心肌组织的3个位点注入共100μl的BMSCs细胞悬液)、注射2组(急性心肌梗死模型大鼠注射同注射1组)、注射3组(正常健康大鼠于相同注射部位注射同注射1组),每组各8只。检测磷酸化p38(p-p38)信号转导蛋白、心肌结蛋白Desmin、肌球蛋白重链MHC和心脏转录因子GATA-4 mRNA的表达。结果模型组和注射1组p-p38蛋白明显高于假手术组和健康对照组(P<0.05);注射1组p-p38蛋白高于模型组、注射2组和注射3组(P<0.05)。Desmin-Brdu和MHC-Brdu双染细胞在注射1组中可见,注射2组和注射3组未见表达。注射3组大鼠GATA-4 mRNA心肌组织中未检出表达,注射1组大鼠心肌组织中GATA-4 mRNA高于注射2组(P<0.05)。注射1组大鼠p-p38与GATA-4 mRNA含量表达呈正相关(P<0.05),而与DesminBrdu、MHC-Brdu无明显相关性。结论急性心肌梗死心血瘀阻证模型大鼠p38信号转导通路激活,可促进移植的BMSCs向心肌细胞方向分化。Objective To study the effect of p38 signaling pathway in cardiac muscle cells of acute myocardial infarction (AMI) rats with heart blood stasis syndrome on differentiation of bone marrow mesenchymal stem cells (BMSCs) . Methods Totally 48 SD rats were randomized into the healthy control group, sham operation group (AMI models) , model group (AMI models with heart blood stasis syndrome), No. 1 injection group (AMI models with heart blood stasis syndrome injected of 100p,1 BMSCs cell suspension) , No. 2 injection group ( AMI models injected of 100p,1 BMSCs cell suspension) and No. 3 injection group (healthy rats injected of 1001xl BMSCs cell sus pension), with 8 in each. The expressions of phosphorylated p38 (p-p38) signal transduction proteins, cardiac Desmin, myosin heavy chain (MHC) and cardiac transcription factor GATA-4 mRNA were detected. Results The expressions of p-p38 proteins in the model group and No. 1 injection group were significantly higher than those in the sham operation group and healthy control group (P 〈 O. 05 ). The expressions of p-p38 proteins in No. 1 injection group were significantly higher than those in the model group, No. 2 injection group and No. 3 injection group (P 〈 0.05). The Desmin-Brdu and MHC-Brdu double-transfected cells were expressed in No. 1 injection group but not expressed in No. 2 injection group and No. 3 injection group. There was no GATA-4 mRNA expression in No. 3 injection group. The expression of GATA-4 mRNA in No. 1 injection group was significantly higher than that in No. 2 injection group (P 〈0. 05). The expression of p-p38 was positively correlated with the expression of GATA-4 mRNA (P 〈 0.05), but with no significant correlation with Desmin-Brdu and MHC-Brdu. Conclusion BMSCs transplantation can induce myocardial cells of AMI rats with heart blood stasis syndrome. It is associated with the activation of p38 signal transduction oathway.
