摘 要:目的通过研究丙泊酚对大鼠大脑睡眠-觉醒相关核团c-fos蛋白表达的影响,探讨丙泊酚诱导睡眠的可能机制。方法选择SD雄性大鼠24只,随机分为四组:生理睡眠组(A组)、生理觉醒组(B组)、生理盐水组(C组)、丙泊酚组(D组),采用免疫组化法测定大鼠睡眠-觉醒相关核团cfos蛋白的表达。结果与A组比较,B组和C组腹外侧视前区(VLPO)的c-fos免疫阳性反应神经fos-IR表达明显减少(P<0.01);结节乳头体核区(TMN)、穹窿周核区(PeF)和外侧下丘脑区(LH)的fos-IR表达明显增加(P<0.01)。与B组比较,D组VLPO区fos-IR表达明显增加,TMN、PeF和LH区fos-IR表达明显减少(P<0.01)。与C组比较,D组VLPO区fos-IR表达明显增加(P<0.01);TMN、PeF和LH区fos-IR表达明显减少(P<0.01)。四组黑质致密部(SNC)区均无fos-IR表达。结论丙泊酚可能通过兴奋睡眠相关核团VLPO区GABA能神经元及抑制觉醒相关核团神经元调节睡眠-觉醒。Objective To study the effect of propofol on c-los protein expression of nuclein related to sleep-awakening in rats brain and investigate the possible mechanism of propofol-indueed sleeping. Methods Twenty-four SD rats were randomly divided into 4 groups: spontaneous sleep group (group A), awaken group (group B), saline group (group C) and propofol group (group D). Immunohistoehemical method was used to measure c-los protein expression of nuclein area related to sleep-awakeing in rat brain. Results Compared with group A, fos-IR was decreased in the ventrolateral preoptic nucleus (VLPO) in groups B and C; while it was increased in the tuberomammillary nucleus (TMN), perifornical nucleus (PeF) and lateral hypothalamic area (LH) (P〈0.01). Compared with group B, fos-IR was increased in the VLPO in group D (P〈0. 01), while it was decreased in the TMN, PeF and LH (P〈0. 01). Compared with group C, fos-IR was increased in the VLPO in group D, while it was decreased in the TMN, PeF and LH (P〈0.01). There was no expression of fos-IR in SNC area in the four groups. Conclusion Propofol moderates sleep-awakening by exciting the GA- BA neurons in the VLPO region of sleep-related nuclein and inhibiting the neurons in awakening-related nuclein.
