HPLC法同时测定盐酸平阳霉素和地塞米松磷酸钠的含量

摘　　要：目的：建立同时测定盐酸平阳霉素和地塞米松磷酸钠含量的方法。方法：采用高效液相色谱法。色谱柱为依利特Hypersil ODS2 C18，流动相为乙腈-水（28：72，v／v），流速为1．0ml／min，检测波长为250nm，柱温为30．0℃，进样量为20μ1。结果：盐酸平阳霉素和地塞米松磷酸钠的检测质量浓度分别在0．03-2．10、0．02-1．10mg／ml范围内与各自峰面积积分值呈良好的线性关系（r=-0．9999、r=0．9997）；精密度、稳定性、重复性试验的RSD〈0．88％；平均加样回收率分别为100．05％、99．81％．RSD分别为0．77％、1．33％（n=9）。结论：该方法简便、快速、准确、灵敏度高，可用于同时测定盐酸平阳霉素和地塞米松磷酸钠的含量。OBJECTIVE： To establish a method for simultaneous determination of bleomycin hydrochloride and dexamethasone sodium phosphate. METHODS： HPLC method was adopted. The determination was performed on Elite Hypersil ODS2 C18 column with mobile phase composed of acetonitrile-water （28 ： 72, V/V） at the flow rate of 1.0 ml/min. The detection wavelength was set at 250 nm and the column temperature was set at 30.0 ℃. The sample size was 20 μ1. RESULTS： The linear range of bleomyin hydro- chloride and dexamethasone sodium phosphate were 0.03-2.10 mg/ml （r=-0.999 9） and 0.02-1.10 mg/ml （r= 0.999 7）, respectively. RSDs of precision, stability and reproducibility tests were all lower than 0.88%. The average recoveries were 100.05% （RSD= 0.77%, n=9） and 99.81% （RSD=l.33%, n=9）. CONCLUSIONS： The method is simple, rapid, accurate and highly sensitive, which can simultaneously determine the contents of bleomycin hydrochloride and dexamethasone sodium phosphate.
