摘 要:目的通过生物荧光法检测耐甲氧西林金黄色葡萄球菌(MRSA)特异性蛋白PBP2a,以达到快速检测MRSA的目的。方法采用生物荧光技术分别对不同浓度下、普通金黄色葡萄球菌标准菌、临床分离的耐甲氧西林葡萄球菌进行检测,以MRSA PBP2a蛋白提取物标准品为对照;通过T-211D激光诱导检测仪软件数据转换导入Excel软件做曲线图。结果不同浓度下PBP2a蛋白提取物标准品,均在横坐标200(T线)处有不同的荧光信号,且随着蛋白浓度的增加,峰值增高;普通金黄色葡萄球菌由于PBP2a蛋白未被荧光信号捕捉到,所以未在200(T线)出现峰值;临床分离的耐甲氧西林葡萄球菌在约200(T线)与标准品均在同一位置捕捉到荧光信号,且荧光信号与MRSA PBP2a蛋白的含量成正比关系。结论在一定浓度下生物荧光技术快速检测到MRSA特异性蛋白PBP2a,从而达到快速检测MRSA的目的,对临床快速诊断有重要参考价值。OBJECTIVE To detect the PBP2a protein in methicillin-resistant Staphylococcus aureus (MRSA) with the use of bioluminescenee assay so as to rapidly detect the MRSA strains. METHODS By means of the biolumines- cence assay, the common standard Staphylococcus aureus strains and the clinical isolates of MRSA strains under different concentrations were detected, then the PBP2a extracted from the MRSA strains were set as controls, and the data were converted and imported to the Excel to make curves by using T-211D laser induced detector. RESULTS All the different concentrations of MRSA-PBP2a protein standards had different fluorescence signals at 200 on the horizontalaxis (T line), besides,there was a increasing trend for peaks with the increasing concentra- tions of protein. As the S. aureus standards did not include MRSA-PBP2a protein, which could not be captured by the fluorescence signals, the peaks did not occur at 200 (T line). The clinical isolates of MRSA and the S. aureus strains have been captured the fluorescence signals at the same place ( around 200, T line ) ~ the fluorescence sig- nals were positively correlated with the content of PBP2aprotein in the MRSA strains. CONCLUSION The biolumi- nescence assay can rapidly detect the PBP2a in the MRSA strains under certain concentrations so as to rapidly de- tect the MRSA strains, which has significant value in the rapid clinical diagnosis.
