月光花豆乙醇提取物对急性肺损伤大鼠的保护作用

摘　　要：目的探讨月光花豆乙醇提取物(CABE)对急性肺损伤(ALI)大鼠的保护作用。方法大鼠尾静脉注射脂多糖5 mg·kg-1复制ALI模型,免疫组化法测定大鼠肺组织中核因子-κBp65(NF-κB p65)的表达,酶联免疫吸附(ELISA)法测定肺组织白细胞介素-1β(IL-1β)的含量,同时观察肺组织病理形态、肺组织的湿干质量比例(W/D)和肺组织髓过氧化物酶(MPO)的活性。结果模型大鼠和CABE治疗大鼠肺组织W/D分别为5.05±0.24,4.35±0.21;IL-1β含量分别为(320.60±19.73),(294.42±13.95)pg·mL-1;MPO活性分别为(1.31±0.18),(1.04±0.18)U·g-1;与模型大鼠比较,CABE治疗大鼠肺组织损伤减轻,IL-1β含量减少,肺组织W/D和MPO活性降低,NF-κB p65表达降低(P<0.01)。结论 CABE对脂多糖诱导的ALI大鼠具有明显的保护作用,干扰NF-κB途径和抑制IL-1β的生成可能是其作用机制之一。Objective To investigate the protective effects of ethanol extract of calonyction acculeatum beans （CABE） on rats with acute lung injury （ALI）. Methods The ALl model was induced via intravenous injection with 5 mg · kg-1 lipopolysaccharide （LPS）. The expression of nuclear factor-κB 1065 （ NF-κB p65 ） was determind by immunohistochemistry （ IHC ）, and the content of interleukin-1β （IL-1β ） in the lung tissue was detected by enzyme-linked immunosorbent assay （ELISA）. Meanwhile,lung tissue histopathology, the wet-dry weight ratio of lung and the activity of myeloperoxidase （MPO） in the lung tissues were observed. Results The wet-dry weight ratio of lung of the model group and CABE treatment group was 5.05 ± 0.24, 4.35 ±0.21, respectively ; the content of IL-113 was （320.60±19.73 ）, （294.42± 13.95 ） pg · mL^-1 , respectively ; the MPO activity was （1.31±0.18 ） , （1.04±0.18） U · g^-1, respectively. Compared with the model group,CABE lessened the lung injury,decreased IL-1β level, reduced the lung dry-wet weight ratio and activity of MPO, and down-regulated the expression of NF-KB/p65 （P〈0.05）. Conclusion CABE has remarkedly protective effect on ALI induced by LPS in rats. One of the mechanisms may be related to reducing the secretion of IL-1β and interfering with the activity of NF-κB.
