摘  要:目的建立测定兔房水中泊沙康唑浓度的高效液相色谱法。方法兔眼局部滴用泊沙康唑后,用高效液相色谱法测定其在兔房水中的浓度。采用Diamonsil C18 为色谱柱;流动相为1%乙酸溶液鄄乙腈(50∶50),柱温25 ℃,流速0. 4 mL/ min,检测波长262 nm。结果在0. 179 -3. 580 μg/ mL 浓度范围内,泊沙康唑峰面积和质量浓度呈良好的线性关系(r =0. 999 8),回收率为99. 38%,日内、日间精密度RSD 均小于3. 0%。结论该方法准确、可靠,可用于测定兔房水中泊沙康唑含量。Objective To establish an HPLC method for determining the content of posaconazole in rabbit aqueous humor. Methods A total of 50 μL of posaconazole eye drops was administered in rabbit eyes. The concentration of posaconazole in rabbit aqueous humor was determined by HPLC. The separation was performed on Diamonsil C18 column, the mobile phase was 1% acetic acid solution-acetonitrile (50:50 ) with the flow rate of 0.4 mL/min, the detection wavelength was at 262 nm and column temperature was 25 ℃. Results The calibration curve of posaconazole was linear within the concentration range of 0. 179 -3. 580μg/mL, r = 0.999 8. The recovery rate was 99.38% and the relative standard deviations (RSD)of intra-day and inter-day assays were less than 3.0%. Conclusion The method is accurate and reliable, and it can be used for the content determination of posaconazole in rabbit aqueous humor.
