摘 要:目的:观察益智仁盐炙对正常大鼠结肠水通道蛋白4(AQP4)的影响,研究益智仁燥性及盐炙润燥的作用机制。方法:将SPF级SD大鼠分为正常动物组、生益智仁组、盐益智仁组、清水炒益智仁组、生益智仁加盐组及生益智仁加增液汤组,各组动物每天灌胃给药1次,连续28天。于第28天处死动物,剖取结肠组织,采用免疫组织化学检测结肠中AQP4的表达情况。结果:与正常动物组比较,生益智仁组结肠AQP4表达显著增加(P<0.01);与生益智组比较,盐益智仁组、清水炒益智仁组、生益智仁加盐组及生益智仁加增液汤组结肠AQP4表达均显著降低(P<0.05或P<0.01),差异均具有统计学意义。结论:益智仁能增加正常大鼠结肠AQP4表达,促进肠道水分重吸收,从而造成便秘,体现其燥性效应;而益智仁盐炙后能降低其引起的燥性效应,其作用机制与盐炙过程中的加热炒制和辅料食盐有关。Objective:To study the effect of Fructus Alpiniae Oxyphyllae (FAO) Stir-frying with Salt Water on AQP4 of normal rats colon, so as to study the mechanism of dry effects and salt-processed moistening-dryness of FAO stir-frying with salt water. Methods:The SPF rats were divided into 6 groups: normal group, crude FAO group, FAO stir-frying with salt water and water groups, crude FAO coupled with salt group, FAO and increased liquid soup combination group. All rats were orally administered once a day for 28 d. All rats were sacrificed on the 28th day, and dissected the colon, then detected the expression of AQP4 of colon by immunohistochemistry. Results:Compared with normal group, the expression of AQP4 of crude FAO group were increased sig- nificantly(P〈~0. 01); compared with crude FAO group, the expression of AQP4 of FAO Stir-frying with salt water and water groups, crude FAO coupled with salt group, FAO and increased liquid soup combination group were decreased significantly(P〈0.05 or P〈0.01). Conclusion: FAO can increase the expression of AQP4 of normal animal colon, to promote intestinal reabsorption of water, then lead to constipation and reflect its dry effects; However, FAO stir-frying with salt water can reduce its dry effects by FAO, the mechanism was related to stir-frying and salt in the process of stir-frying with salt water.
