摘  要:从发病鸡的粪便中分离出具有多重耐药性的CHNⅣ型铜绿假单胞菌,测定黏菌素的最小抑菌浓度(MIC)和抗生素后效应(PAE),以4×MIC黏菌素作用临床分离株,除药后分别继续培养0、1、2 h,无菌水代替黏菌素设为空白组。检测细菌细胞膜完整性,巨噬细胞吞噬细菌数量和细菌对Caco-2细胞的黏附力。与空白组相比,黏菌素作用后细菌细胞膜发生严重损坏,被巨噬细胞吞噬的细菌数量明显增多,细菌对Caco-2的黏附力显著下降。上述指标随着细菌除药后培养时间的延长而改变。说明黏菌素对鸡源性铜绿假单胞菌存在PAE,且可能与细菌细胞膜被破坏,细菌对细胞黏附力下降和巨噬细胞对其吞噬能力增强有关。Multi-drug-resistant Pseudomonas aeruginosa (P.aeruginosa)CHN IV was isolated from excrements of infected chickens. To determine colistin's minimum inhibitory concertration (MIC)and post-antibiotic effect (PAE),respectively, the 4×MIC colistin was treated with CHN IV ,the bacterial liquids were continuously cultured for 0 h, 1 h and 2 h after agent removed. A blank group was treated with sterile water in place of colistin, Cell membrane integrity of P.aeruginosa CHN IV, number of bacteria phagocytosed by macrophages,and bacterial adhesion on Caco-2 cells in each group were observed. Compared with control group, there were serious damage in bacterial cell membranes, significant increase in the number of bacteria phagocytosed by macrophages (P〈0.01), and notable decrease in the bacterial adhesion on Caco-2 cells in other three groups (P〈0.01). The results showed that colistin had PAE on chicken-derived P.aeruginosa. The mechanism may be related to injury of bacterial cell membranes, decrease of the bacterial adhesion on cells, as well as intensification of the phagocytosis of macrophages.
