摘  要:为了研究烟草(Nicotiana tabacum L.)叶片表皮蜡质组成以及蜡质的含量,以NC89为材料,对其叶片进行蜡质提取,并利用气相色谱-质谱联用仪(GC-MS)和气相色谱-火焰离子化检测仪(GC-FID)对提取的蜡质进行定性和定量分析。同时采用扫描电镜(SEM)观察其叶片表面蜡质超微结构。结果表明:烟草叶表皮蜡质经GC-MS分析,共鉴定出化合物23种,主要为直链烷烃与支链烷烃,并含有少量的初级醇以及三萜类化合物;表皮蜡质含量为(5.578±0.463)μg/cm2,直链烷烃[(2.717±0.345)μg/cm2]与支链烷烃[(2.052±0.120)μg/cm2]为蜡质主要组分。直链C31烷烃含量最高,初级醇和三萜类化合物含量相对较低。扫描电镜观察到烟草叶片上下表面蜡质层都较平滑,无明显的蜡质晶体。In order to study the composition and content of tobacco (Nicotiana tabacum L. ) leaf cutic ular waxes, tobacco leaf of cuhivar NC89 was used as the plant material to extract the cuticular waxes, the wax composition and content were qualitatively and quantitatively analyzed by gas chromatography-mass spectrometry (GC-MS) and gas chromatography-flame ionization detector (GC-FID). At the same time, the wax uhrastructure of tobacco leaf surface was observed by scanning electron microscope . The results showed that 23 chemical compounds were identified by GC-MS analysis, the main components of cuticualr waxes were n-alkanes and branched-alkanes, with small amount of primary alcohols and triterpenoids. The total content of cuticular waxes was [-(5. 578±0. 463)μg/cm2, dominated by n-alkanes [(2. 717±0.345) μg/cm2], and branched-alkanes [-(2. 052±0. 120) μg/cm2]. The composition of n-hentriacontane was the highest, primary alcohols and triterpenoids were rela tively low. Cuticular wax layer on the upper and lower surface of tobacco leaf was even, smooth andwithout visible wax crystals.
