摘  要:目的检测HCA587蛋白疫苗在小鼠体内诱导的抗体水平,并分析抗体效价与抗肿瘤作用的相关性。方法按照不同的免疫方案以液体形式尾根部皮下注射给小鼠,将肿瘤细胞在小鼠左侧胁腹部皮下接种以建立肿瘤模型。用酶联免疫吸附实验(ELISA)方法检测抗体总IgG或IgG各亚型的水平,并用游标卡尺测量肿瘤大小。结果 HCA587蛋白疫苗诱导的抗HCA587抗体总IgG效价高于1∶1 024×104,其水平与肿瘤大小呈显著负相关。γ-干扰素(IFN-γ)敲除后,该疫苗的抗肿瘤效果完全消失,同时抗HCA587的IgG2a亚型效价显著下降。结论 HCA587蛋白疫苗诱导的IgG抗体效价很高,具有一定的抗肿瘤作用,其中IgG2a可能与抗肿瘤关系密切。HCA587, also known as MAGE-C2, is a novel cancer-testis antigen highly expressed in a number of malignancies, making it a promising target for tumor immunotherapy. In this study, we aimed to detect the antibody level induced by HCA587 protein vaccine, and analyze the correlation between the titer of antibody induced by the vaccine and the anti-tumor effect. Mice were immunized with different vaccination strategies by subcutaneous injections at tail root. In tumor model, mice were inoculated with tumor cells by subcutaneous injection on the flank. Two weeks after the second immune vaccination, the level of HCA587-specific IgG or IgG isotypes was detected using enzyme-linked immunosorbent assay (ELISA). Moreover, tumor volume was measured with a caliper every 2-3 days. Our data showed HCA587 protein vaccine could induce high level of HCA587- specific total IgG antibody, with titers more than 1:1 024×10^4, which was negatively correlated with tumor volume. In IFN-γ knock out (KO) mice, the anti-tumor effect induced by HCA587 protein vaccine completely disappeared, and HCA587-specific IgG2a isotype decreased significantly compared with wild type mice. These results suggest that the HCA587 protein vaccine can induce high titer HCA587-specific antibody, which could resist tumor growth in mice. And IgG2a isotype may play a major role in the anti-tumor effect.
