周期性压力对兔软骨细胞整合素α3β1的影响

摘　　要：观察周期性压力对兔关节软骨细胞增殖及其分泌整合素α3β1的影响。通过体外分离培养胎兔关节软骨细胞,取第3代细胞接种于6孔板,分别设置无干预对照组(A组)、周期性压力组(B组压力25PSI,频率2Hz),持续性压力组(C组压力25PSI,持续加压)。压力干预每天2 h,连续干预3 d后,以WST-1细胞增殖及细胞毒性检测试剂盒检测各组软骨细胞增殖情况(OD值),并以ELISA法检测各组培养上清中整合素α3β1含量。细胞OD值A组为1.16±0.08,B组为1.52±0.12,C组为1.36±0.13,三组间两两比较OD值差异均有统计学意义(P<0.05)。A组整合素α3β1水平为3.64±0.15 pg/ml,B组为4.03±0.19 pg/ml,C组为3.77±0.14 pg/ml,三组间两两比较整合素α3β1差异有统计学意义(P<0.05)。周期性压力刺激兔软骨细胞,能增加软骨细胞的增殖作用和整合素的含量,其作用比持续性压力更强。To observe the effect of cyclic pressure on proliferation of rabbit articular chondrocytes and integrin-α3β1 production. The chondrocytes were isolated from fetal rabbit articular cartilage and cultured in vitro. Third-generation chondrocytes were plated in culture plates. We set the no intervention control group（ group A）,cyclicpressure group（ 25PSI,group B）,persistent pressure group（ group C,25PSI,sustained pressure）. 2 h a day of continuous pressure intervention,after 3 d,the proliferation of chondrocytes in each group were detected by the cytotoxic WST-1 cell proliferation assay kit（ OD value）,and ELISA assay was used to measure the culture supernatant of integrin α 3 β 1 levels. The OD value of group A was 1. 16 ± 0. 08,group B was 1. 52 ± 0. 12,and group C was 1. 36 ± 0. 13. Comparison of OD value of each group,the difference had statistical significance（ P〈0. 05）,group A of integrin α 3 β 1 level was 3. 64 ± 0. 15 pg/ml,group B was 4. 03 ± 0. 19 pg/ml,group C was 3. 77 ± 0. 14 pg/ml,comparison of integrin α 3 β 1 of each group,the difference was statistically significant（ P〈0. 05）. Cyclic pressure stimulation of rabbit chondrocytes in cartilage can increase cell proliferation and integrin level. Cyclic pressure is more effective than continuous pressure.
