摘  要:目的:观察阻断老年大鼠心肌细胞过氧化物酶体增殖物激活型受体α(PPARα)信号通路对白细胞介素-1β(IL-1β)表达水平的影响。方法:将培养的老年大鼠心肌细胞分为空白对照组、溶剂对照组、阿托伐他汀组、PPAR拮抗剂GW6471+阿托伐他汀组。分别加入细胞培养液、二甲基亚砜(DMSO)、阿托伐他汀、阿托伐他汀+PPARα拮抗剂GW6471。用RTPCR和western blot方法检测各组细胞IL-1βmRNA和IL-1β蛋白含量。结果:①与空白对照组相比,溶剂对照组大鼠心肌细胞IL-1βmRNA表达水平和蛋白含量均无显著差异(P>0.05);②与空白对照组相比,阿托伐他汀组IL-1βmRNA表达水平和蛋白含量均显著降低(P<0.01);③GW6471+阿托伐他汀组IL-1βmRNA表达水平及蛋白含量均显著高于阿托伐他汀组(P<0.05),但仍低于空白对照组(P<0.05)。结论:阿托伐他汀抑制老年大鼠心肌细胞IL-1β表达的作用可能与其激活PPARα信号通路有关。Objective: To investigate the changes of Interleukin-1β (IL-1β) expression in myocytes of aging rats after blockage of signal pathway of peroxisome proliferator activated receptor α (PPARα). Methods: Primary cultures of cadiocyte were got from aging rats. Cadiocyte were divided into control group, dimethyl sulfoxide (DMSO) group, atorvastatin group, and atorvastatin plus GW6471 group, which treated respectively by cell culture medium, DMSO, atorvastatin, and atorvastatin plus GW6471. The expression of IL-1β mRNA was detected by RT--PCR, and content of IL--1β protein was determined by Western blot. Results: There was no significant difference between control and DMSO groups in expression level of IL- 1β mRNA and protein (P〉0. 05). The expression levels of IL-- 1β mRNA and protein in atorvastatin group were significantly lower than those in control group (P〈0.01). Both the mRNA and protein expressions of IL--1β in atorvastatin plus GW6471 group were significantly higher than those in atorvastatin group (P〈0.05), but still lower than those in control group (P〈0.05). Conclusion: The atorvastatin inhibits the IL-1β expression by activating the PPARα signal pathway in myocytes of aging rat.
