摘 要:通过溶胀-锚定(swelling-anchoring)法,用含刚性疏水胆固醇侧基的磷酰胆碱基二元无规共聚物(PMC)构筑稳定两性离子表面涂层,并以含柔性疏水十二烷基酯侧基的磷酰胆碱基二元无规共聚物(PML)为参照聚合物。以水接触角为考察指标对浸涂工艺进行优化。结果发现,丙酮中溶胀5 s后,迅速在质量分数为0.2%PMC73聚合物溶液中浸涂2 h,所得磷酰胆碱两性离子聚合物涂层s-PMC-PC的亲水性最佳,XPS测试结果表明,表面N和P含量分别为2.50%和2.72%。该涂层经过100℃加热2 h和空气中放置2个月处理,接触角基本不变,说明涂层表面稳定性优异;经过75%乙醇中浸泡6 h,1%SDS超声15 min处理,接触角虽然有所升高,但仍远低于对照聚合物涂层,说明该涂层界面附着力有所提高。s-PMC-PC涂层表面抵抗非特异性蛋白质吸附性能优异,Fg和BSA蛋白吸附量可分别降低91.5%和90.8%。A stable zwitterionic surface was fabricated by a simple swelling-anchoring method with phosphorylcholine-based amphiphilic copolymer (PMC) containing rigid hydrophobic cholesteryl side group.The phosphorylcholine-based amphiphilic copolymer (PML) containing flexible alkyl side group was used as control.The influence of types of swelling solvents,swelling time and dip-coating time on the hydrophilicity of the modified PC plate was studied.The results showed that the optimal hydrophilicity could be achieved under the following conditions:acetone as swelling agent,5s of swelling time,0.2wt% of amphiphilic copolymer and 2hof dip-coating time.XPS test results showed that the surface N and P contents of the coating surface were 2.50% and 2.72% respectively.When the coating was heated at 100℃ for 2h or stored in the air for 2 months,the contact angle had no changes.After the coating was soaked in 75% ethanol for 6 h or by 1% SDS ultrasonic for 15 min,although the contact angle was increased,s-PMC-PC had still relatively better surface stability and improved interfacial stability.The Fg and BSA protein adsorption on the s-PMC-PC surface had been reduced by 91.5% and 91.5% respectively.
