摘　　要：拟通过建立血管性痴呆(vascular dementia,Vad)患者永生化淋巴细胞的实验,探索在不同实验室条件下有效建立患者永生细胞库的方法。采用EB病毒体外转化外周血B淋巴细胞技术建立永生细胞株;采用RT-PCR技术和染色体G显带技术对其进行质量鉴定。研究中共采集了15例患者血液样本,最终建成3个永生细胞株,其余12株在平均培养至18天时发生退化。分析结果证明B淋巴细胞在被EB病毒转化后核型不会发生改变。转化过程中在有些情况下会存在一个危机期,成功渡过此期的细胞可无限增殖,具体出现危机期的机制及如何有效渡过危机期的实验条件还有待进一步研究。Investigation of optimized methods for the establishment of B lymphoblastoid cell lines by establishing immortalized human lymphocytes cell lines of vascular dementia patients.EB virus transformation technology of peripheral blood B lymphocytes was used to establish the permanent lymphoblastoid cell lines of vascular dementia patients.RT-PCR technique was used to detect the expression of Epstein Barr virus encoded latent membrane protein 1 （LMP-1） gene.Chromosome karyotype analysis was carried out on the transformation of the cell lines.15 patients blood samples were collected,and eventually 3 immortal cell lines were established,and the remaining 12 samples turned degradation from the 18th day.After being transformed by EB virus,the chromosome karyotype of B lymphocytes didn＇t change.There was a crisis in the process of transformation,the cells which successfully got through this period could have unlimited proliferation.However,the mechanism and the experimental conditions to get through the crisis remains to be further studied.
