摘 要:为了解二斑叶螨(Tetranychus urticae)几丁质合成酶(CHS)基因结构及其潜在应用,采用同源基因克隆技术克隆二斑叶螨几丁质合成酶基因TuCHS,克隆获得的目的基因全长4 608bp,编码1 535个氨基酸,其蛋白预测分子质量约为175.48ku,理论等电点6.92。其包含EDR和QRRRW 2个几丁质合成酶特有的标签序列,18个保守基序。ExPASy在线分析表明,该基因具有18个跨膜螺旋、1个氨基化位点、5个糖基化位点以及12个酰基化位点。结合其他物种CHS基因构建系统发育树,结果表明,该基因催化区域与其他3种昆虫CHS1基因的相似性为80%-89%,属于几丁质合成酶A类基因。In order to study gene structure and its potential application,chitin synthase gene of Tetranychus urticae,TuCHS,was cloned by homology gene cloning techniques.The full-length of TuCHSgene contained 4 608bp encoding 1 535amino acids.The putative protein of TuCHSgene has predicted molecular mass of 175.48ku with a theoretical pI of 6.92.It included two specific tag sequences of chitin synthase:EDR,QRRRW and eighteen conserved motifs.The online ExPASy analysis showed that the gene had 18transmembrane helices,one Amidation site,5N-glycosylation sites and 12N-myristoylation sites.The phylogenetic tree of TuCHSgene has been constructed combined with those from other species.The results showed that the identities of the gene catalytic domain with those from the other three insects were from 80%-89%,which belongs to the A class of chitin synthase gene.
