摘 要:采用PCR-SSCP技术对贵州黑山羊和贵州白山羊GHRL基因进行单核苷酸多态性检测,并分析其多态与体重、体尺性状间的关联性。结果发现,在GHRL基因外显子4处存在1个同义突变位点(C345T),表现为2种基因型,分别命名为CC和CT。基因型与体重、体尺性状关联分析结果显示,在贵州黑山羊(麦坪)群体中,CT基因型个体的体重、体高和胸围均显著高于CC基因型个体,而在贵州黑山羊(纳雍)和贵州白山羊群体中差异不显著。研究结果揭示,GHRL基因突变位点(C345T)影响贵州黑山羊生长性状可能性较大,该位点有望作为山羊生长性状的一个标记辅助选择位点。PCR-SSCP was used to detect the single nucleotide polymorphism of GHRL gene of Guizhou Black Goat and Guizhou White Goat, and its corelation with body weight and body size traits was analyzed. The result showed that a SNP(C345T) was detected in Exon 4 of GHRL gene, which was a sense mutation and led to two genotypes named CC and CT. The study of relationships between genotype and body weight and body size traits revealed that the body weight, body height and chest girth of individuals with genotype CT were very significantly better than genotype CC in Guizhou Black Goat from Maiping Country. But no significant difference between other indexes and goats(Guizhou Black Goat from Nayong Country and Guizhou White Goat) was found. The consequence indicated that the SNP of GHRL gene could be regarded as a candidate molecular marker of MAS affecting growth traits in Guizhou Black Goat.
