摘  要:【目的】观察电针对血管性痴呆(vascular dementia,VD)模型大鼠海马CA1区β淀粉样蛋白(Aβ1-40)基因与蛋白表达的影响。【方法】选用SD大鼠,随机分为假手术组,模型组,电针组(电针百会、大椎穴),西药组(吡拉西坦,剂量为0.8 mg·kg-1·d-1);后3组采用改良二血管阻断(2-VO)法复制VD模型,并运用Morris水迷宫检测各组大鼠学习记忆能力,采用Western blot法检测Aβ1-40蛋白表达,荧光定量PCR法检测Aβ1-40 mRNA表达。【结果】模型组第1~4天逃避潜伏期均显著延长,停留于原平台象限总时间及单次进入原平台的时间均显著减少,海马CA1区Aβ1-40 mRNA和蛋白表达均显著增加(P〈0.05或P〈0.01);电针组可显著缩短各时段逃避潜伏期,增加停留于原平台象限总时间及单次进入原平台时间,显著抑制海马CA1区Aβ1-40 mRNA与蛋白表达(均P〈0.05或P〈0.01)。【结论】电针改善VD大鼠学习记忆能力与其能抑制VD模型大鼠海马CA1区Aβ1-40 mRNA与蛋白表达有关。Objective To observe the effect of electro-acupuncture(EA) on learning and memory abilities as well as the protein and gene expression of amyloid protein β1-40(Aβ1-40) in the hippocampal CA1 area of vascular dementia( VD) rats. Methods SD rats were randomized into sham operation group, model group, EA group(EA on Baihui and Dazhui points) and western medicine group(treated with Piracetam 0.8 mg·kg-1· d-1). Rats except for the sham operation group were modeled by modified two-vessel occlusion(2-VO). Morris water maze test was employed to study the learning and memory behavior of rats. The protein expression of Aβ1-40 was measured by Western blotting, and its gene expression was measured by fluorescence quantitative PCR. Results On experimental day 1-4, escape latency of rats in the model group was prolonged, total time for staying in the original quadrant and average time for staying in the original quadrant were shortened, and Aβ1-40 protein and gene expression levels in the hippocampal CA1 area were increased(P0.05 or P0.01). In EA group, escape latency was shortened, total time for staying in the original quadrant and average time for staying in the original quadrant were prolonged, and Aβ1-40 protein and gene expression levels in the hippocampal CA1 area were decreased( P〈 0.05 or P〈0.01). Conclusion EA is effective on improving the learning and memory abilities of VD rats, and the mechanism is probably related with the regulation of Aβ1-40 protein and gene expression in the hippocampal CA1 area of VD rats.
