摘 要:【目的】探讨补肾化瘀法抗光老化对基质金属蛋白酶MMP-1与MMP-3表达的影响。【方法】采用8-甲氧沙林(8-MOP)/长波紫外线照射SD大鼠法,复制光老化大鼠模型,并对该动物模型给予补肾化瘀中药干预,连续15 d,采用免疫组织化学SP法,分别检测空白对照组、补肾化瘀中药干预组和光老化模型组大鼠皮肤MMP-1、MMP-3的表达,使用图像分析软件分析各组照片的单位灰度值并进行比较,根据灰度值的大小判断MMP-1、MMP-3的表达水平。【结果】建立了光老化大鼠模型。经过免疫组织化学染色显示,补肾化瘀中药干预组大鼠皮肤切片中阳性染色颗粒明显减少,MMP-1和MMP-3表达显著减弱。使用图像分析软件分析结果显示,光老化模型组大鼠皮肤中MMP-1和MMP-3表达显著高于空白对照组,差异有统计学意义(P<0.05);中药干预组大鼠皮肤中MMP-1和MMP-3表达水平显著降低,与模型对照组比较,差异有统计学意义(P<0.05)。【结论】补肾化瘀中药能显著降低光老化模型大鼠皮肤中MMP-1和MMP-3的表达水平,促进光老化大鼠皮肤损害的修复,具有一定的抗光老化作用。Objective To study the influence of anti-photoaging through replenishing kidney and dissolving blood stasis on the expression of matrix metalloproteinase 1(MMP-1) and MMP-3. Methods SD rat skin photoaging model was established by irradiation with 8-methoxypsoralen/ultraviolet A(UVA) at 320-400 nm. The animal models were given intervention of Chinese medicine with the actions of replenishing kidney and dissolving blood stasis for 15 continuous days. Immunohistochemistry S-P method was adopted for the detection of MMP-1 and MMP-3 expression in the rat skin of the blank control group, Chinese medicine intervention group and photoaging model group. By using image analysis software, we analyzed and compared the mean grey value of the photographs of each group, and evaluated the expression levels of MMP-1 and MMP-3. Results The photoaging rat model has been established successfully. The results of immunohistochemical staining showed that positivestaining particles in the rat skin of Chinese medicine intervention group were decreased significantly. The results of image analysis software analysis showed that the expression levels of MMP-1 and MMP-3 in photoaging model group were significantly higher than those in the blank control group( P〈0.05), and then were decreased after intervention by Chinese medicine, the differences being statistically significant as compared with the photoaging model group(P〈0.05). Conclusion Chinese medicine with the actions of replenishing kidney and dissolving blood stasis exerts certain anti-photoaging action on photoaging model rat, and its mechanism is probably related with the decrease of MMP-1 and MMP-3 expression levels, and with the enhancement of skin lesion repair.
