摘 要:【目的】观察四物汤对环磷酰胺所致贫血小鼠肝细胞凋亡及凋亡执行分子Caspase3表达的影响。【方法】选用48只C57BL/6J小鼠随机分为空白组,模型组,四物汤低、中、高剂量组(剂量分别为5、10、20 g·kg-1·d-1),肌苷(阳性对照,剂量为0.06 g·kg-1·d-1)组。以腹腔注射环磷酰胺(剂量为0.1 g/kg)复制贫血模型,采用TUNEL法检测肝细胞凋亡,Western blot法检测Caspase-3表达。【结果】四物汤可显著升高贫血模型小鼠外周血红细胞计数、白细胞计数以及血红蛋白含量,显著降低肝细胞凋亡百分率,下调Caspase3蛋白表达(均P﹤0.05)。【结论】四物汤补益肝血的作用机理可能与其减少肝细胞凋亡有关。Objective To investigate the effects of Siwu Decoction(SD) on hepatocyte apoptosis and Caspase 3 expression in mice with anemia induced by cyclophosphamide. Methods Forty-eight C57BL / 6J mice were randomly divided into 6 groups, namely blank control group, model control group, carnine group( positive control group, treated with 0.06 g·kg-1·d-1carnine), and low-, middle- and high-dose SD groups(in the dosage of 5, 10, and 20 g·kg-1·d-1, respectively). The anemia model was established by intraperitoneal injection of cyclophosphamide(in the dosage of 0.1 g / kg). TUNEL assay was carried out to detect hepatocyte apoptosis and Western blotting method was applied to assess the protein expression of Caspase 3. Results SD had obvious effect on increasing the count of erythrocytes, leukocytes and hemoglobin in peripheral blood of anemia model mice, and could also decrease the apoptotic rate of hepatocytes and down-regulate the expression of Caspase 3(P0.05). Conclusion The mechanism of SD for reinforcing liver and nourishing blood may be associated with its reduction of hepatocyte apoptosis.
