摘  要:【目的】基于转录组测序技术研究当归与5种重要经济作物的序列相似性,为当归栽培、鉴定、加工、配伍及开发提供理论支持。【方法】岷县现场采集当归样品,并经专家鉴定为当归品种[Angelica sinensis(oliv.)diels]。采用RNA plant Plus方法提取岷归头、岷归尾的总RNA,质量鉴定合格后构建测序文库。测序文库检测合格后在Illummina HiSeq 2000测序仪上完成双端测序。采用相关生物信息学分析软件,实现研究目标。【结果】通过转录组测序技术,获取了63 585个当归表达序列标签(EST),并在UniProt(date:2012.09)数据库中注释了30 432个当归基因,均分布于939种物种;其中序列分布频率较高的前5种物种分别为葡萄、蓖麻、毛果杨、大豆、苜蓿属。【结论】已注释基因主要分布于目前研究较深入的5种重要经济植物,成为当归制剂开发的分子生物学基础。Objective To explore the sequence similarity of Angelica sinensis( Oliv.) Diels and five kinds of economic crops, thus to provide genetic evidence for the cultivation, identification, processing and herbal combination of A. sinensis. Methods Samples of A. sinensis were collected in Min County of Gansu Province, and were confirmed as Angelica sinensis(Oliv.) Diels by relevant expert. The total RNA of the heads and the tails of A. sinensis were extracted by RNA plantPlus kit. The sequencing library of RNA was constructed and qualified by RNeasy Micro kit, and then the qualified sequencing library was checked by ILLummina HiSeq 2000 device. Finally, relevant data were mined by CLC Genomics Workbench( version 5.5) and UniProt( date: 2012.09). Results The results of transcriptome sequencing showed that 63 585 unigenes have been obtained from A. Sinensis, which constructed the genomic library of A. Sinensis. And then 30 432 unigenes were annotated in UniProt library. These genes were extensively distributed in 939 species, and the top five species having higher distribution frequency were Vitis vinifera, Ricinus communis, Populus trichocarpa, Glycine max and Medicago truncatula. Conclusion We have primarily explored that the genomic resources of A. Sinensis are mainly distributed in five kinds of economic crops, which will supply the data of molecular biology for the development of A. Sinensis preparations.
