摘 要:目的建立一种快速、准确测定人血清中甲氨蝶呤浓度的高效液相色谱法。方法色谱柱为HypersilC。流动相为甲醇一乙腈一磷酸盐缓冲液(2:7:91),流速为0.8mL·min-1,检测波长313nm;血清样品上SPE柱,以含甲醇一乙腈一磷酸盐缓冲液(50:50:250)为洗脱剂,收集洗脱液直接进样测定。结果甲氨蝶呤血清样品线性范围为0.03—50.00μg·mL~,2组低、中、高不同浓度(A组:3.41,6.82,27.27μg·mL~;B组:0.05,0.21,0.85μg-mL。)方法回收率均大于97%。日内、日间RSD均小于5%。结论本方法准确性好、灵敏度高、操作简便,完全可满足儿科临床甲氨蝶呤血药浓度监测需要。Objective To establish for determination the concentration Methods Chromatographic column a rapid and accurate HPLC method of methotrexate in human serum. was Hypersil Cl8 column; mobile phase was methanol,acetonitrile and phosphate buffer (2:7:91 ), a flow rate was 0. 8 mL · min- 1 ; detection wavelength was 313 nm. Serum sam- ple was injected into SPE column. Methanol/acetonitrile/phosphate buff- er (50: 50: 250) was used as the eluent, and the collection of eluent was detected directly. Results The linear range of methotrexate in serum samples was 0. 03 - 50. 00 μg · mL- t, coefficient of recoveries of low, medium and high concentrations in two groups ( group A : 3.41, 6. 82, 27.27 μg· mL-1 ; group B : 0. 05, O. 21, 0. 85 μg · mL- 1 ) were higher than 97%. Inter day and inner day RSD were less than 5%. Conclusion This method has good accuracy, high sensitivity, convenient opera- tion. It can fully satisfy the requirement of concentration monitoring of methotrexate in the pediatric clinical practice.
